Animal models are considered essential in research ensuing elucidation of human disease processes and subsequently, testing of potential therapeutic strategies. This is especially true for neurodegenerative disorders, in which the first steps in pathogenesis are often not accessible in human patients. Alzheimer's disease is vastly becoming a major medical and socioeconomic problem in our aging society. Valid animal models for this uniquely human condition should exhibit histopathological, biochemical, cognitive, and behavioral alterations observed in Alzheimer's disease patients. Major progress has been made since the understanding of the genetic basis of Alzheimer's disease and the development and improvement of transgenic mouse models. All present Alzheimer's disease models developed are partial but nevertheless essential in further unraveling the nature and spatial and temporal development of the complex molecular pathology underlying this condition. One of the more recent transgenic attempts to model Alzheimer's disease is the APP23 transgenic mouse. This article describes the development and assessment of this human amyloid precursor protein overexpression model. We summarize histopathological and biochemical, cognitive and behavioral observations made in heterozygous APP23 mice, thereby emphasizing the model's contribution to clarification of neurodegenerative disease mechanisms. In addition, the first therapeutic interventions in the APP23 model are included.  相似文献

In ‘a secular age’ (Taylor 2007), pastoral care is no longer exclusively associated with specific religious traditions and communities. Pastoral caregivers who work in secular institutions provide care to religious and nonreligious people alike, and in several Western societies the term pastoral care is used in relation to nonreligious (humanist) care. In secular contexts, the term ‘pastoral care’ is often replaced by the term ‘spiritual care.’ Spiritual care, however, is provided by various professionals, so pastoral caregivers face the challenge of developing adequate and convincing language to explain what is distinctive about their work. In this article, the authors turn to philosophical language in order to develop a conceptual understanding of pastoral care that does not depend on the specific worldview—religious or nonreligious—of either pastoral caregivers or receivers of pastoral care. Using the work of Taylor (1989, 2007) and Murdoch (1970), we explain pastoral care as engaging with people’s attempts to orient in ‘moral space’ and the distinctive quality of pastoral care as ‘representing the Good.’ Murdoch associates ‘the Good’ with a secular idea of transcendence that is both a movement beyond the ego and an engagement with the reality of human vulnerability, suffering, and evil. We argue that pastoral caregivers who ‘represent the Good’ have the task not only of supporting the existential and spiritual processes of individuals but also of promoting dialogue and social justice and of critiquing dehumanizing practices in the organizations in which they work and in society at large.  相似文献

Objective: Both the distressed (Type D) personality (i.e. the combination of negative affectivity and social inhibition traits) and dysfunctional parenting styles are associated with anxiety and depression. As parenting styles have been related to personality development, dysfunctional parenting styles may also be associated with Type D personality. We examined whether remembered parenting was associated with anxiety and depression in cardiac patients and whether Type D personality mediated this relationship.

Methods: Our sample comprised 435 patients treated with percutaneous coronary intervention (PCI) and 123 patients with congestive heart failure (CHF). Patients completed the Hospital Anxiety and Depression Scale, Type D Scale (DS14), and Remembered Relationship with Parents (RRP10) scale.

Results: Remembered parenting was significantly associated with higher anxiety and depression levels and Type D personality. In multivariable linear regression analyses, Type D personality accounted for 25–29% of the variance in anxiety and 23–46% of the variance in depression, while remembered parenting was no longer significantly associated with these domains. Sobel tests and bootstrapping indicated that Type D personality mediated the relationship between remembered parenting and anxiety and depression.

Conclusion: Type D personality mediated the relationship between remembered parenting and anxiety and depression in both PCI and CHF patients.  相似文献

There are a number of differences between normative and non-normative systemic therapy. It is useful to understand normative thinking because that is a traditional orientation. Non-normative clinical work also has much to offer both philosophically and practically: It is characterized by the premise that people's behavior makes sense given their histories and current context. Therefore, when understood from the perspective of the individual, even his or her psychopathology would make sense. Using a composite case analyzed in normative terms and treated non-normatively, the author illustrates how to bridge these two approaches. Solution-focused methods constitute the interventions of choice.  相似文献

The article deals with the perceptions of Judaism of Savonarola's most important predecessors, the Dominican preacher Giovanni Dominici andthe Franciscan preacher Bernardino da Siena, who were active in fifteenth century Florence. These preachers offer two distinct models of mendicant preaching regarding the Jews: Dominici based his views on theological arguments, while Bernardino focused on contemporary Jewish moneylenders and their destructive dangers. The preachers' opposition to the Jews was grounded in their political ideal of Florentine republic as a purified sacred body where there was no place for polluting outsiders as the Jews.Both preachers succeeded in influencing the fate of the Jews. Dominici might have had a hand in the banishment of Jews from Venice and there is circumstantial and linguistic evidence that he was responsible for the change in Florentine policy against the Jews in the legislation of 1406. Bernardino caused the banishment of Jews from many Italian towns and was responsible for measures taken against them, such as segregation. Both preachers were successful in keeping the Jews out of the city of Florence, although they failed in their attempt to havethe Jews expelled from the Florentine territories. To search for genetic influence on human fertility differentials appears inconsistent with past empirical research and prior interpretations of Fisher's fundamental theorem of natural selection. We discuss Fisher's theorem and give reasons why genetic influences may indeed account for individual differences in human fertility. We review recent empirical studies showing genetic influence on variance in fertility outcomes and precursors to fertility. Further, some of the genetic variance underlying fertility outcomes overlaps with that underlying fertility precursors. Findings from different cultures, different times, different levels of data, and both behavioral and molecular genetic designs lead to the same conclusion: Fertility differentials are genetically influenced, and at least part of the influence derives from behavioral precursors that are under volitional control, which are themselves genetically mediated.  相似文献

In this study we examined the factor structure of the Child Behavior Checklist (CBCL) filled out by group care workers. Group care workers' judgements were collected on 846 children and adolescents treated in various residential settings in The Netherlands. Using confirmatory factor analysis, we were able to show that the original CBCL factor model based on parental judgments of child behavior also fits for the judgments of group care workers. This means that the well known 8 narrow-band syndromes (Withdrawn, Somatic Complaints, etc.) as well as the 2 broad-band syndromes (Internalizing and Externalizing) can be used to interpret the CBCL scores of group care workers. This confirmation of the CBCL factor structure is a first step to add a group care worker version to the CBCL family. However, as a second step, normative data need to be gathered to further enhance the use of the CBCL for group care workers.  相似文献

The present studies examined whether differences in need for cognitive closure (NCC) were related to differences in regulatory control when confronted with authority. In two studies, levels of regulatory control were measured when participants resisted (Study 1; N = 46) or prepared to resist the influence attempt of an authority figure (Study 2; N = 50). Results showed that resisting the influence attempt from a high‐authority figure was more depleting for participants higher in NCC compared to individuals lower in NCC. However, when they were given instructions and time to prepare the act of resistance, individuals high in NCC actually showed an increase in regulatory control. Authority is usually viewed as a general principle of influence; however, the present studies suggest that there are individual differences that influence how people may experience interactions with authorities.  相似文献

In this study, the construct validity and reliability of the Empowerment questionnaire (EMPO) that was developed in Dutch youth care was examined. The 12-item EMPO focuses on measuring parental empowerment in raising their children. The three components of psychological empowerment (intrapersonal, interactional, and behavioral) form the rationale for the EMPO. Non-clinical (n?=?673) and clinical (n?=?1,212) data were used. Construct validity was tested by the factorial structure, measurement invariance, correlations with other instruments (PSQ-S and SDQ), and empowerment differences between the two groups. Reliability was determined by testing the internal consistency and test-retest reliability. The results show that the factorial validity of the EMPO was sufficient to good, the EMPO was measurement invariant for various subgroups, and the EMPO scales were negatively correlated with parenting stress (PSQ-S) and child behavioral problems scales (SDQ). Furthermore, the clinical group was less empowered, and the correlation between parental empowerment and child behavioral problems was stronger in this group. In addition, both groups of parents had relatively high scores on the interactional component. In the clinical group, however, parental scores on the interactional component were less correlated with scores on the intrapersonal and behavioral components. Finally, reliability analyses showed a largely sufficient to good internal consistency and test-retest reliability. The EMPO seems to be an instrument with sufficient to good construct validity and reliability. Further research is recommended regarding the underlying assumptions, other aspects of validity, representativeness, and the way it should be used as a tool by professionals for supporting parental empowerment.  相似文献
