Apolipoprotein receptors belong to an evolutionarily conserved surface receptor family that has intimate roles in the modulation of synaptic plasticity and is necessary for proper hippocampal-dependent memory formation. The known lipoprotein receptor ligand Reelin is important for normal synaptic plasticity, dendritic morphology, and cognitive function; however, the in vivo effect of enhanced Reelin signaling on cognitive function and synaptic plasticity in wild-type mice is unknown. The present studies test the hypothesis that in vivo enhancement of Reelin signaling can alter synaptic plasticity and ultimately influence processes of learning and memory. Purified recombinant Reelin was injected bilaterally into the ventricles of wild-type mice. We demonstrate that a single in vivo injection of Reelin increased activation of adaptor protein Disabled-1 and cAMP-response element binding protein after 15 min. These changes correlated with increased dendritic spine density, increased hippocampal CA1 long-term potentiation (LTP), and enhanced performance in associative and spatial learning and memory. The present study suggests that an acute elevation of in vivo Reelin can have long-term effects on synaptic function and cognitive ability in wild-type mice.  相似文献

General cognitive ability (GCA) is a recognized construct for predicting job performance and capacity to learn. However, it has recently been argued that the time constraints under which GCA is assessed might provoke test anxiety, which negatively biases GCA scores. This can then lead to erroneous rejection of qualified candidates in personnel hiring contexts. This paper aimed to investigate: (1) to what extent candidates’ GCA scores increase when tested without time constraints and the ability of this GCA score to predict job performance; and (2) the personality characteristics that hinder GCA test performance under time constraints. Results from two field studies conducted in an actual personnel selection context partially confirmed the hypotheses. They revealed that, aside from the improvement of all candidates’ GCA scores when time constraint was removed, only GCA assessed without time constraints predicts job performance. Furthermore, while all candidates’ scores were influenced by the time constraint condition, individuals who are anxious, low-impulse, low value-questioning and deliberating are more penalized by the time constraint condition of such testing and, thus, are more likely to be erroneously eliminated in a selection process.  相似文献

Nociceptin/orphanin FQ (N/OFQ) peptide and its receptor are not only ubiquitously expressed in mammalian brain and spinal cord but are also abundant in limbic structures, particularly in the hippocampus. The widespread distribution of N/OFQ reflects the broad spectrum of its biological actions such as nociception, food intake, spontaneous locomotor activity, and learning and memory processes. Since the hippocampus is involved in the control of adrenocortical activity, its role in stress-related phenomena is well characterized. In male Wistar rats, we first examined the effects of acute restraint stress (120?min) on the brain immunohistochemical localization of N/OFQ. The analysis carried out on sections obtained at the onset of stress revealed enhanced expression of N/OFQ in CA1, CA3, and the dentate gyrus as well as increased plasma corticosterone concentrations. Next, we examined whether endogenous glucocorticoid hormone plays a role in the modulation of hippocampal N/OFQ expression in response to stress. To this end, rats were injected with corticosterone (1?mg/kg) or subjected to restraint stress 1 week after adrenalectomy. Two hours after corticosterone administration, plasma glucocorticoid concentrations were comparable to those observed after restraint stress, while N/OFQ expression had significantly increased in all the hippocampal subfields examined. By contrast, in adrenalectomized rats, stress did not modify protein expression. These results confirm that stress can affect N/OFQ expression and that glucocorticoids may constitute hormonal mediators of this complex interplay.  相似文献

This article aims to verify whether the empirical data presented in the literature on sexually assaulted victims support the hypothesis advanced by various aetiological models, which is that cognitions play an important role in the development and maintenance of Post-Traumatic Stress Disorder. The first objective is to present state-of-the-art results on sexually assaulted victims' post-traumatic cognitions and their link to post-assault symptomatology. Eighty blindfolded healthy female subjects participated in an active and a passive straight-ahead pointing task to study the estimation of the subjective sagittal middle in the presence or absence of an active haptic exploration. Subjects were to point straight-ahead with their left or right index finger starting from different right- or left-sided locations, while performing either an active movement or with the hand being passively moved by the examiner. Results showed: (i) a significant effect of the hand used only on the active straight-ahead pointing task performance; (ii) a significant effect of the starting position both on the active and passive pointing performance; (iii) a significant correlation between the two protocols only in terms of spatial bias (algebraic error) but not in terms of precision (absolute errors). These results are discussed with regards to normal and neuropsychological studies of the egocentric frame of reference.  相似文献

Two experiments examined the effects of reductions in cortical cholinergic function on performance of a novel task that allowed for the simultaneous assessment of attention to a visual stimulus and memory for that stimulus over a variable delay within the same test session. In the first experiment, infusions of the muscarinic receptor antagonist scopolamine into the medial prefrontal cortex (mPFC) produced many omissions but did not impair rats' ability to correctly detect a brief visual stimulus. However, these animals were highly impaired in remembering the location of that stimulus following a delay period, although in a delay-independent manner. In the second experiment, another group of animals with selective 192IgG-saporin lesions of the nucleus basalis magnocellularis (nBM) were not impaired under conditions of low-attentional demand. However, when the stimulus duration was reduced, a significant memory impairment was observed, but similar to the results of the first experiment, the nBM-lesioned animals were not impaired in attentional accuracy, although aspects of attention were compromised (e.g., omissions). These findings demonstrate that (1) cortical cholinergic depletion produces dissociable deficits in attention and memory, depending on the task demands, (2) delay-independent mnemonic deficits produced by scopolamine are probably due to impairments other than simple inattention, and (3) working memory deficits are not simply dependent on attentional difficulties per se. Together, these findings implicate the nBM cortical cholinergic system in both attentional and mnemonic processing.  相似文献

Sixty-five right- and left-handed preschool and school children were tested on three reach-to-grasp tasks of different levels of complexity, performed in three space locations. Our goal was to evaluate how the effect of attentional information related to object location interacts with task complexity and degree of handedness on children's hand selection. Results revealed a shift to the non-preferred hand in the contralateral hemispace, which was more or less pronounced according to the level of task complexity. The subject's degree of handedness also influenced this shift, since strongly lateralized children exhibited a greater use of their preferred hand than less lateralized ones for actions in the contralateral hemispace. These findings confirm that hand selection is to some extent adaptable to task demand and environmental context.  相似文献

Many laboratory studies have demonstrated an age effect on episodic memory which is in contrast with the preservation of semantic memory. The aim of this study was the assessment of age effects on autobiographical memory according to the length of the retention interval, taking into account the episodic and semantic components. A total of 52 subjects, aged between 40 and 79, were divided into four age groups. They were tested with a sophisticated autobiographical questionnaire consisting of two tasks, one involving the recall of semantic information and another the recall of episodic events. Results revealed that episodic recall deteriorated more with age and retention interval than semantic recall. These data, gathered using an ecological test, confirm age differences demonstrated by laboratory tests on the episodic-semantic distinction. Furthermore, the profile of results obtained for the recall of specific detailed events, and analysed according to age of encoding, confirms the distribution of episodic memories across the lifespan, as modelled by Rubin, Wetzler, and Nebes (1986) with the cue-word technique.  相似文献

Whether or not conscious recollection in autobiographical memory is affected in schizophrenia is unknown. The aim of this study was to address this issue using an experiential approach. An autobiographical memory enquiry was used in combination with the Remember/Know procedure. Twenty-two patients with schizophrenia and 22 normal subjects were asked to recall specific autobiographical memories from four lifetime periods and to indicate the subjective states of awareness associated with the recall of what happened, when and where. They gave Remember, Know or Guess responses according to whether they recalled these aspects of the event on the basis of conscious recollection, simply knowing, or guessing. Results showed that the frequency and consistency of Remember responses was significantly lower in patients than in comparison subjects. In contrast, the frequency of Know responses was not significantly different, whereas the frequency of patients' Guess responses was significantly enhanced. It is concluded that the frequency and consistency of conscious recollection in autobiographical memory is reduced in patients with schizophrenia.  相似文献
