Working memory deficits in transgenic rats overexpressing human adenosine A2A receptors in the brain

Adenosine receptors in the central nervous system have been implicated in the modulation of different behavioural patterns and cognitive functions although the specific role of A(2A) receptor (A(2A)R) subtype in learning and memory is still unclear. In the present work we establish a novel transgenic rat strain, TGR(NSEhA2A), overexpressing adenosine A(2A)Rs mainly in the cerebral cortex, the hippocampal formation, and the cerebellum. Thereafter, we explore the relevance of this A(2A)Rs overexpression for learning and memory function. Animals were behaviourally assessed in several learning and memory tasks (6-arms radial tunnel maze, T-maze, object recognition, and several Morris water maze paradigms) and other tests for spontaneous motor activity (open field, hexagonal tunnel maze) and anxiety (plus maze) as modification of these behaviours may interfere with the assessment of cognitive function. Neither motor performance and emotional/anxious-like behaviours were altered by overexpression of A(2A)Rs. TGR(NSEhA2A) showed normal hippocampal-dependent learning of spatial reference memory. However, they presented working memory deficits as detected by performance of constant errors in the blind arms of the 6 arm radial tunnel maze, reduced recognition of a novel object and a lack of learning improvement over four trials on the same day which was not observed over consecutive days in a repeated acquisition paradigm in the Morris water maze. Given the interdependence between adenosinic and dopaminergic function, the present results render the novel TGR(NSEhA2A) as a putative animal model for the working memory deficits and cognitive disruptions related to overstimulation of cortical A(2A)Rs or to dopaminergic prefrontal dysfunction as seen in schizophrenic or Parkinson's disease patients. 相似文献

Recent work shows that the inability to inhibit basic motor responses (like pressing a button) is related to overeating, weight gain and overweight. In the present study it was tested whether this inability to inhibit motor responses - or impulsivity - can differentiate between successful and unsuccessful restrained eaters. A typical preload and food exposure paradigm was used and it was hypothesized that only the high-restrained eaters that are simultaneously inefficient inhibitors of prepotent motor responses would overeat when confronted with tempting foods. In line with the hypothesis, the data show that overeating follows from an interaction between restraint and impulsivity; high-restrained eaters only overate when they were also impulsive. It is concluded that being restrained per se is not a determinant of overeating. Being a restrained eater only bears the risk of overeating in case of coexisting impulsivity. 相似文献

It has been proposed that the uniqueness of human cumulative culture may be attributable to humans' greater orientation toward copying the process of behavior (imitation), as compared with the products (emulation), resulting in particularly high fidelity transmission. Following from previous work indicating that adult human participants can exhibit cumulative learning on the basis of product copying alone, we now investigate whether such learning involves high fidelity transmission. Eighty adult human (Homo sapiens) participants were presented with a task previously shown to elicit cumulative learning under experimental conditions, which involved building a tower from spaghetti and modeling clay. Each participant was shown two completed towers, ostensibly built by previous participants, but actually built to prespecified designs by the experimenter. This end state information was provided either in the form of photographs, or the presence of actual towers. High fidelity matching to these end states was apparent in both demonstration conditions, even for a design that was demonstrably suboptimal with regard to the goal of the task (maximizing tower height). We conclude that, although high fidelity transmission is likely to be implicated in cumulative culture, action copying is not always necessary for this to occur. Furthermore, since chimpanzees apparently copy behavioral processes and well as products, and also transmit behavior with high fidelity, the stark absence of unequivocal examples of cumulative culture in nonhumans may be attributable to factors other than imitative ability. 相似文献

The purpose of this study was to explore spirituality and its relationship to resilience for women in late life. Over thirty interviews with six women aged 80 and older provide a dataset allowing for the phenomenological investigation of spiritual resilience. Themes emerged illustrating the components of spiritual resilience. The components of spiritual resilience are having divine support, maintaining purpose, and expressing gratitude. These factors are essential to the women’s resilience and act as mechanisms that promote high levels of subjective well-being and an overall good quality of life. Studies of transmission biases in social learning have greatly informed our understanding of how behaviour patterns may diffuse through animal populations, yet within-species inter-individual variation in social information use has received little attention and remains poorly understood. We have addressed this question by examining individual performances across multiple experiments with the same population of primates. We compiled a dataset spanning 16 social learning studies (26 experimental conditions) carried out at the same study site over a 12-year period, incorporating a total of 167 chimpanzees. We applied a binary scoring system to code each participant’s performance in each study according to whether they demonstrated evidence of using social information from conspecifics to solve the experimental task or not (Social Information Score—‘SIS’). Bayesian binomial mixed effects models were then used to estimate the extent to which individual differences influenced SIS, together with any effects of sex, rearing history, age, prior involvement in research and task type on SIS. An estimate of repeatability found that approximately half of the variance in SIS was accounted for by individual identity, indicating that individual differences play a critical role in the social learning behaviour of chimpanzees. According to the model that best fit the data, females were, depending on their rearing history, 15–24% more likely to use social information to solve experimental tasks than males. However, there was no strong evidence of an effect of age or research experience, and pedigree records indicated that SIS was not a strongly heritable trait.
