Excitation of the mesocorticolimbic pathway, originating from dopaminergic neurons in the ventral tegmental area (VTA), may be important for the development of exaggerated fear responding. Among the forebrain regions innervated by this pathway, the amygdala is an essential component of the neural circuitry of conditioned fear. The functional role of the dopaminergic pathway connecting the VTA to the basolateral amygdala (BLA) in fear and anxiety has received little attention. In vivo microdialysis was performed to measure dopamine levels in the BLA of Wistar rats that received the dopamine D(2) agonist quinpirole (1 μg/0.2 μl) into the VTA and were subjected to a fear conditioning test using a light as the conditioned stimulus (CS). The effects of intra-BLA injections of the D(1) antagonist SCH 23390 (1 and 2 μg/0.2 μl) and D(2) antagonist sulpiride (1 and 2 μg/0.2 μl) on fear-potentiated startle (FPS) to a light-CS were also assessed. Locomotor performance was evaluated by use of open-field and rotarod tests. Freezing and increased dopamine levels in the BLA in response to the CS were both inhibited by intra-VTA quinpirole. Whereas intra-BLA SCH 23390 did not affect FPS, intra-BLA sulpiride (2 μg) inhibited FPS. Sulpiride's ability to decrease FPS cannot be attributed to nonspecific effects because this drug did not affect motor performance. These findings indicate that the dopamine D(2) receptor pathway connecting the ventral tegmental area and the basolateral amygdala modulates fear and anxiety and may be a novel pharmacological target for the treatment of anxiety. 相似文献

Pharmacotherapy follow-up (PTF) is the professional practice in which the pharmacist takes charge of the needs of the patients related to their drugs. OBJECTIVE: To validate a questionnaire to measure the ASE psychosocial determinants (attitude, social influence, self-efficacy) of PTF and the Prochaska & DiClemente's stage of change in Spanish community pharmacists. METHOD: Postal survey to 482 Asturian community pharmacists (test and retest). Validity was estimated in 134 pharmacists answering the test. Stability was estimated in 85 pharmacists answering the test and retest. Exploratory factorial analysis was used to investigate construct validity and produced 4 factors and 104 items. Those factors had high reliability (Cronbach's a between 0,87 and 0,96). DISCRIMINANT VALIDITY: Pharmacists in extreme stages of change showed significant differences of attitude, motivation and self-efficacy. Although we are far from a universally accepted pattern of impaired function at altitude, there is evidence indicating motor, perceptual, memory and behavioural deficits in adults. Even relatively low altitudes (2500 m) may delay reaction time, and impair motor function. Extreme altitude exposure (>5000 m) may result in more pronounced impairment that can persist after returning to the lowlands. Research into the effects of altitude exposure earlier in development is lacking by comparison. Un-acclimatized children can suffer from acute mountain sickness, and, in native populations born at altitude, subtle cognitive and behavioural deficits suggest incomplete adaptation to hypoxia. The study of neurobehavioural functioning at altitude may provide important information about the effects of clinical hypoxia on the human brain and behavioural development. 相似文献

Hemi-field visual-spatial attention was studied on a timed letter cancellation task in proficient readers from 7th to 9th grades. Participants maintained greater accuracy in identifying targets in the left quadrants (top/bottom) when compared with the right quadrants (top/bottom), revealing a leftward bias in their responses. Effects of gender and age were evident on performance; females scored significantly higher than males, and 9th graders scored significantly higher than 7th and 8th graders. Thus, a substantial shift in brain maturation may take place from 8th to 9th grades or from the ages of 13.0 to 14.11. Discriminative characteristics of the task are analyzed by assigning greater value to targets located in the top left quadrant. The 9th graders, as compared to the younger students, and the girls, as compared to the boys, evidenced a reading-direction-related bias toward systematic preference for the upper-left to lower-right quadrants. 相似文献

Recognition of words and kaleidoscope pictures showed a double dissociation of left and right hippocampal activity using magnetic source imaging (MSI). MSI has advantages over alternative imaging techniques that measure hemodynamic changes for identifying regional changes in brain activity in real time and on an individual subject basis without the need for image subtraction. In this study, lists of words or kaleidoscope pictures were presented for memorization followed by tests of list items and foils during which brain activity was recorded. There was greater activation in the left than the right hippocampus with abstract nouns (e.g., relief) and greater activation in the right than the left hippocampus with kaleidoscope pictures. This dissociation was evident on a case by case basis. This study demonstrates the specialization of the two medial temporal lobe (MTL) regions, including the hippocampi, for mnemonic processing of verbal and pictorial items that are difficult to encode verbally. 相似文献

The question I address in this article is whether it is morally wrong for a lawyer to represent a client whose purpose is immoral or unjust. My answer to this question is that it is wrong, prima facie. This conclusion holds, even accepting certain traditional principles of lawyer's professional ethics, such as the right of defence and the so‐called principle of ‘adversarial’ litigation. Both the adversarial system and the right of defence are sufficient to support or justify the right of potential clients (and citizens in general) to defend their interests in the judicial system and to do so with the technical assistance of a lawyer. This right includes a right to pursue unjust or immoral purposes (within the law). However, having a right to do X does not mean that it is morally permissible to do X. We can have a right to do something morally wrong. This being so, the fundamental moral reason for a lawyer not to accept representation for a client with an immoral purpose is that it is, prima facie, morally wrong to help someone do something wrong. 相似文献
