Rodent models have been especially useful for investigating adolescent ethanol exposure. However, there is a paucity of studies examining sex differences in behavioral intoxication from adolescent ethanol drinking. Here, we used an ethanol drinking model to investigate if adolescent rats of both sexes readily drink ethanol to measurable behavioral intoxication, indicated by increased impulsive action and motor incoordination. Beginning on postnatal day (P) 28, male and female Long‐Evans rats were given 30‐min access to a solution of sucrose (20%) or sweetened ethanol (20% sucrose +15% ethanol) every other day until P60 and once after 2 weeks of forced abstinence (on P75). On alternate (nondrinking) days, rats were reinforced with a food pellet for making a cued nosepoke response. Beginning on P56, rats were tested in this task after drinking sessions to assess ethanol‐induced changes in impulsive action, defined as premature responding prior to cue presentation. Motor coordination was assessed before and after drinking sessions using an incline plane test. Adolescent male and female rats readily consumed ethanol to behavioral intoxication, measured as reduced motor coordination. Following forced abstinence, females displayed greater ethanol‐induced impulsive action. These studies provide evidence for sex differences in behavioral intoxication following adolescent ethanol drinking.  相似文献

Rats' operant responding for sucrose rewards in the 1st half of a session can vary directly with the conditions of reward in the 2nd half. The authors investigated whether that induction effect represented an animal model of emotive states. Rats pressed a lever for either ethanol or sucrose rewards in the 1st half of a 40-min session. The reward in the 2nd half of the session was, across conditions, 1% sucrose, a food pellet, or the same reinforcer delivered in the 1st half. When subjects responded for sucrose, upcoming reward type had little influence on responding in the 1st half of the session. However, when subjects responded for ethanol, upcoming 1% sucrose and food-pellet reinforcement both produced increases in responding. The results suggest that the procedure produced different emotive states, but further work is needed to support such a model. The results also have potential applied implications.  相似文献

Previous research suggested that allocation of responses on concurrent schedules of wheel‐running reinforcement was less sensitive to schedule differences than typically observed with more conventional reinforcers. To assess this possibility, 16 female Long Evans rats were exposed to concurrent FR FR schedules of reinforcement and the schedule value on one alternative was systematically increased. In one condition, the reinforcer on both alternatives was .1 ml of 7.5% sucrose solution; in the other, it was a 30‐s opportunity to run in a wheel. Results showed that the average ratio at which greater than 90% of responses were allocated to the unchanged alternative was higher with wheel‐running reinforcement. As the ratio requirement was initially increased, responding strongly shifted toward the unchanged alternative with sucrose, but not with wheel running. Instead, responding initially increased on both alternatives, then subsequently shifted toward the unchanged alternative. Furthermore, changeover responses as a percentage of total responses decreased with sucrose, but not wheel‐running reinforcement. Finally, for some animals, responding on the increasing ratio alternative decreased as the ratio requirement increased, but then stopped and did not decline with further increments. The implications of these results for theories of choice are discussed.  相似文献

Experiments examined the effects of acute doses of chlordiazepoxide upon ethanol self-administration in the rat. A concurrent-schedule procedure was used that employed choice between ethanol (5%) and a second fluid (either water or a 1% sucrose solution). When ethanol and water were the available fluids, chlordiazepoxide at doses of 15 and 20 mg/kg reduced ethanol-reinforced responding and intake, with a greater reduction occurring at the 20 mg/kg dose. However, when ethanol and sucrose were concurrently available, in many rats only the 20 mg/kg dose of chlordiazepoxide reduced ethanol-reinforced responding. The differences in dose response function occurred in most animals without large changes in the baseline ethanol-reinforced responding across the two concurrent conditions. Thus the dose-effect curve relating chlordiazepoxide and ethanol self-administration can be altered, dependent upon the nature of the concurrently available reinforcers.  相似文献

These experiments examined the own-price and cross-price elasticities of a drug (ethanol mixed with 10% sucrose) and a nondrug (10% sucrose) reinforcer. Rats were presented with ethanol-sucrose and sucrose, both available on concurrent independent variable-ratio (VR) 8 schedules of reinforcement. In Experiment 1, the variable ratio for the ethanol mix was systematically raised to 10, 12, 14, 16, 20, and 30, while the variable ratio for sucrose remained at 8. Five of the 6 rats increased ethanol-reinforced responding at some of the increments and defended baseline levels of ethanol intake. However, the rats eventually ceased ethanol-reinforced responding at the highest variable ratios. Sucrose-reinforced responding was not systematically affected by the changes in variable ratio for ethanol mix. In Experiment 2, the variable ratio for sucrose was systematically increased while the ethanol-sucrose response requirement remained constant. The rats decreased sucrose-reinforced responding and increased ethanol-sucrose-reinforced responding, resulting in a two- to 10-fold increase in ethanol intake. Experiment 3 examined the substitutability of qualitatively identical reinforcers: 10% sucrose versus 10% sucrose. Increases in variable-ratio requirements at the preferred lever resulted in a switch in lever preference. Experiment 4 examined whether 10% ethanol mix substituted for 5% ethanol mix, with increasing variable-ratio requirements of the 5% ethanol. All rats eventually responded predominantly for the 10% ethanol mix, but total amount of ethanol consumed per session did not systematically change. In Experiment 5, the variable-ratio requirements for both ethanol and sucrose were simultaneously raised to VR 120; 7 of 8 rats increased ethanol-reinforced responding while decreasing sucrose-reinforced responding. These data suggest that, within this ethanol-induction procedure and within certain parameters, demand for ethanol-sucrose was relatively inelastic, and sucrose consumption was independent of ethanol-sucrose consumption. Demand for sucrose, on the other hand, was relatively elastic, and ethanol-sucrose readily substituted for it. The results are discussed in terms of applying a behavioral economic approach to relationships between drug and nondrug reinforcers.  相似文献

Nicotine functions as a negative feature in a Pavlovian discriminated goal-tracking task. Whether withholding of responding to the conditional stimulus (CS) reflects nicotine functioning as a conditioned inhibitor is unknown. Accordingly, the present research sought to determine whether nicotine trained as a negative feature passed the retardation-of-acquisition and summation tests, thus characterizing it as a pharmacological (interoceptive) conditioned inhibitor. In the retardation test, rats received either nicotine (0.4 mg/kg) or chlordiazepoxide (5 mg/kg) negative feature training in which the drug state signaled when a 15-sec light CS would not be paired with sucrose; light was paired with sucrose on intermixed saline sessions. Following acquisition of the discrimination, both groups received nicotine CS training in which sucrose was intermittently available on nicotine but not intermixed saline sessions. Acquisition of conditioned responding to the nicotine CS was slower in the nicotine negative feature group than in the chlordiazepoxide negative feature group. In the summation test, rats were assigned to either the nicotine negative feature group or a pseudoconditioning control. In this control, the light CS was paired with sucrose on half the nicotine and half the saline sessions. Both groups also received excitatory training in which a white noise CS was paired with sucrose. The summation test consisted of presenting the white noise in conjunction with nicotine. Conditioned responding evoked by the white noise was decreased in the negative feature but not the pseudoconditioning group. Combined, the results provide the first evidence that an interoceptive stimulus (nicotine) can become a conditioned inhibitor.  相似文献

Research has demonstrated that rats' rates of operant behavior maintained by 1% sucrose reinforcement in the first half of an experimental session are heightened when food-pellet reinforcers, rather than 1% sucrose, will be available in the second half. Experiment 1 showed that rats that had been displaying this positive induction effect acquired a new response more quickly when 1% sucrose was used to reinforce the novel response than did rats that had not been displaying induction. Experiment 2 showed that this enhanced acquisition remained even when the new response was learned in a new environment. Experiment 3 showed that unconditioned rates of making a new response did not differ between subjects that had or had not been displaying induction. Experiment 4 further showed that significantly different rates of responding on a novel task were not observed when that response was reinforced with a new, non-sucrose reinforcer. Together, the present results suggest that induction results in and/or from an increase in the reinforcing value of the 1% sucrose. As such, the present results have both theoretical and practical implications.  相似文献

In a split-litter design, equal numbers of male and female rats were exposed either to carbon dioxide for 1 min, or to the control condition on either postnatal Days 9-10 or 17-18. At 120 days of age half the numbers of rats in each condition were given subclinical dosages of lithium (1.5 mEq/kg) and pilocarpine (15 mg/kg) or saline. 40 days later all rats were tested in a conditioned fear paradigm. A four-way analysis of variance as a function of the variables anoxia, age at the time of anoxia sex, and subclinical seizure induction indicated a significant interaction between sex and anoxic history. Post hoc analysis showed that performance of male rats exposed to the anoxic condition did not differ from normal or anoxic female rats. The results suggest that only 1 min. of preweaning anoxia may produce permanent changes in contextual fear conditioning for male but not female rats.  相似文献

In the present experiment, sex differences in hemispheric asymmetry during classical conditioning to emotional stimuli are reported. 125 subjects (62 females and 63 males) were shown a slide of a happy face in the right (or left) visual half field (VHF), and simultaneously a slide of an angry face in the left (or right) VHF. Eight groups were formed by the combination of male and female subjects; left and right VHF positions of the angry/happy faces; and the administration/omission of the shock unconditioned stimulus (UCS). Dependent measures were skin conductance responses recorded from both hands. The results during extinction showed a significant larger SCR magnitude to the shock compared to the no-shock groups only for the female subjects. CS position during conditioning was also important in revealing differential responding to either the happy or angry faces. A right hemisphere effect was found for the angry face CS for both the male and female subjects, however with a greater difference for the females.  相似文献

Forty-four male and 44 female subjects, divided on the basis of their need for achievement scores, responded to three short verbal cues in which the central character was either male or female. The three cues (store clerk, social worker, and author) were chosen in order to maximize status differences and to minimize masculinity-femininity differences. In responding to each cue, subjects had to imagine themselves in each role. subjects rated each character on semantic differential scales to provide impressions of personality. Ratings were found to be a function of sex of the subject and need for achievement level, as well as status of the occupation. Previous research reported that rats responding for 1% liquid-sucrose reinforcement when 32% sucrose reinforcement is upcoming will decrease their response rate (contrast) if licking is the dependent measure and increase their response rate (induction) if pressing a lever is the dependent measure. The present study investigated whether induction could be observed when licking served as the dependent measure and whether induction in lever pressing and contrast in licking behaviour could be concurrently observed. Experiment 1 found induction when rats licked to earn the rewards but consumed them at a location separate from the spout licked to earn them. Experiment 2 also found induction when rats earned (and consumed) rewards by licking the same spout throughout the session. Experiment 3 separately measured instrumental lever pressing for sucrose rewards and licking the sucrose during the reward period. We found that both measures increased for 1% sucrose when 32% sucrose reinforcement was upcoming. The present results indicate that the type of response is not the sole determinant of whether contrast or induction is observed. Rather, they suggest that other procedural details, such as the location of reinforcer delivery, influence which effect is observed. The results also indicate that associative processes underlie the appearance of induction in responding for 1% sucrose.  相似文献

Several researchers have suggested that conditioning history may have long-term effects on fixed-interval performances of rats. To test this idea and to identify possible factors involved in temporal control development, groups of rats initially were exposed to different reinforcement schedules: continuous, fixed-time, and random-interval. Afterwards, half of the rats in each group were studied on a fixed-interval 30-s schedule of reinforcement and the other half on a fixed-interval 90-s schedule of reinforcement. No evidence of long-term effects attributable to conditioning history on either response output or response patterning was found; history effects were transitory. Different tendencies in trajectory across sessions were observed for measures of early and late responding within the interreinforcer interval, suggesting that temporal control is the result of two separate processes: one involved in response output and the other in time allocation of responding and not responding.  相似文献

Two experiments examined the processes underlying the suppression of instrumental behaviours by lithium in rats, as reported by Meachum (1988 and this issue). Experiment 1 examined whether presenting a novel sucrose solution prior to lithium chloride administration would overshadow aversion learning to either the stimuli of the operant chamber or to familiar food pellets. After lever pressing had been established, and in the absence of responding, animals received free deliveries of a novel sucrose solution, familiar food pellets, or both, or they were exposed to only the cues of the operant chamber, prior to lithium injections. Lever pressing for food pellets was then assessed. It was found that the animals receiving the novel sucrose, either alone or with the familiar food pellets, pressed more for pellets than either the group receiving only food pellets or the group exposed to only the context. In addition, there was no appreciable difference in the response rates between the context-only group and the group that received the familiar food pellets. These outcomes were interpreted in terms of the novel sucrose overshadowing aversion learning to the context. Experiment 2 investigated whether in fact aversive contextual conditioning could be obtained using the present parameters. This was accomplished by directly manipulating the contexts. In this experiment animals were trained to lever press in two distinctive contexts. Subsequently, one context was paired with the novel sucrose, and the second was experienced in the absence of reinforcement prior to toxicosis. During a subsequent non-reinforced test it was found that responding in the context paired with the novel sucrose was considerably higher than responding in the context that was experienced alone. These findings stand in contrast to the taste-mediated contextual potentiation observed when a consumatory response is used to assess aversive contextual conditioning.  相似文献

Two experiments investigated performance of instrumental lever pressing by rats following post-conditioning devaluation of the sucrose reinforcer produced by establishing an aversion to it. In Experiment I rats responded less in an extinction test after being averted from the sucrose following training on a ratio schedule, but not following an equivalent amount of training on an interval schedule. This was true even though the devalued sucrose would not act as an effective reinforcer on either the ratio or interval schedule. Experiment II provided a further investigation of the insensitivity of interval responding to reinforcer devaluation by comparing test performance under simple extinction with responding when the devalued reinforcer was presented on either a response-contingent or non-contingent schedule during the test. Once again simple extinction performance was unaffected by prior reinforcer devaluation. Furthermore, neither non-contingent nor contingent presentations of the devalued reinforcer significantly depressed responding below the level seen in the extinction condition. Ratio, but not interval performance appears to be controlled by knowledge about the instrumental contingency that encodes specific properties of the training reinforcer.  相似文献

Six male albino rats were placed in running wheels and exposed to a fixed-interval 30-s schedule of lever pressing that produced either a drop of sucrose solution or the opportunity to run for a fixed duration as reinforcers. Each reinforcer type was signaled by a different stimulus. In Experiment 1, the duration of running was held constant at 15 s while the concentration of sucrose solution was varied across values of 0, 2.5. 5, 10, and 15%. As concentration decreased, postreinforcement pause duration increased and local rates decreased in the presence of the stimulus signaling sucrose. Consequently, the difference between responding in the presence of stimuli signaling wheel-running and sucrose reinforcers diminished, and at 2.5%, response functions for the two reinforcers were similar. In Experiment 2, the concentration of sucrose solution was held constant at 15% while the duration of the opportunity to run was first varied across values of 15, 45, and 90 s then subsequently across values of 5, 10, and 15 s. As run duration increased, postreinforcement pause duration in the presence of the wheel-running stimulus increased and local rates increased then decreased. In summary, inhibitory aftereffects of previous reinforcers occurred when both sucrose concentration and run duration varied; changes in responding were attributable to changes in the excitatory value of the stimuli signaling the two reinforcers.  相似文献

Through associative learning, cues for biologically significant reinforcers such as food may gain access to mental representations of those reinforcers. Here, we used devaluation procedures, behavioral assessment of hedonic taste-reactivity responses, and measurement of immediate-early gene (IEG) expression to show that a cue for food engages behavior and brain activity related to sensory and hedonic processing of that food. Rats first received a tone paired with intraoral infusion of sucrose. Then, in the absence of the tone, the value of sucrose was reduced (Devalue group) by pairing sucrose with lithium chloride (LiCl), or maintained (Maintain group) by presenting sucrose and LiCl unpaired. Finally, taste-reactivity responses to the tone were assessed in the absence of sucrose. Devalue rats showed high levels of aversive responses and minimal appetitive responses, whereas Maintain rats exhibited substantial appetitive responding but little aversive responding. Control rats that had not received tone-sucrose pairings did not display either class of behaviors. Devalue rats showed greater FOS expression than Maintain rats in several brain regions implicated in devaluation task performance and the display of aversive responses, including the basolateral amygdala, orbitofrontal cortex, gustatory cortex (GC), and the posterior accumbens shell (ACBs), whereas the opposite pattern was found in the anterior ACBs. Both Devalue and Maintain rats showed greater FOS expression than control rats in amygdala central nucleus, GC, and both subregions of ACBs. Thus, through associative learning, auditory cues for food gained access to neural processing in several brain regions importantly involved in the processing of taste memory information.  相似文献

Children ranging in age from 5 1/2 to 9 years old selected either a male or a female doll in response to questions about who would be “better” at an occupation. The occupations were selected so that half were female and half were male sex-typed occupations. The responses indicated that children have clear sex-typed expectations concerning occupational competence. While the differences between the “female” and “male” occupations were significant at each age level, the interaction revealed an increasing adherence to the stereotypes with increasing age. A sex of child main effect was produced by the tendency of female children to select the female doll more often than did male children. The results are interpreted as demonstrating that children's perceptions of these occupations reflect an evaluative bias and not simply a recognition of adult stereotypes or actual rates of employment in the positions.  相似文献

In Study I measures of cortical thickness were taken in 90-day-old male and female Sprague-Dawley, Wistar, and Long-Evans rats that were born to pregnant females shipped to the laboratory in late pregnancy and that were either gonadectomized at birth or left intact. No evidence for hemispheric differences in cortical thickness was found in any of the groups. Gonadectomy resulted in increased cortical thickness in male Sprague-Dawley and Wistar rats and in small increases in thickness of the left hemisphere in Long-Evans rats, especially males. No effects of gonadectomy were found in females of any strain. The lack of the expected right-over-left difference in cortical thickness in intact males may have been due to prenatal stress caused by housing, handling, and shipping. In Study 2 measures of cortical thickness were taken in 90-day-old male and female Sprague-Dawley rats that were bred in our laboratory, but otherwise similarly treated at birth. In this second study, the expected right-over-left hemispheric difference in cortical thickness was found in intact male rats; none was found in gonadectomized males or in either of the female groups. The lack of asymmetry in gonadectomized males appeared to be due to an increase in thickness in the left hemisphere. Taken together these studies provide support for the idea that hormones of testicular origin suppress cortical enlargement in the perinatal period, particularly in the left hemisphere.  相似文献
