To study the learning performance of pentylenetetrazol- and amygdala-kindled Wistar rats we used the following learning tests: short-term memory was tested in the response-to-change model, brightness discrimination was tested in a Y-chamber, and two-way active avoidance learning was tested in a shuttle-box. Short-term memory was not impaired by both kindling procedures. Considering two-way active avoidance learning the performance of pentylenetetrazol (PTZ)-kindled rats was significantly diminished. This effect persists over a period of 4 weeks. However, amygdala (AMY)-kindled rats acquired this task like the controls. In brightness discrimination reaction (BDR) the learning performance of PTZ-kindled animals was not influenced. Although the acquisition of BDR was nearly identical, the 24-h retention was remarkably diminished in AMY-kindled rats. It was hypothesized that the different kindling procedures interfere in different ways and extent with neuronal circuits resulting in different functional impairments.  相似文献

Two experiments were conducted with rats. The first study showed that in the T-maze a brightness discrimination learning problem was more difficult than a spatial learning problem, but in a Ross-maze a brightness problem was less difficult than a spatial task. T-maze brightness and Ross-maze spatial tasks were found to be of equal difficulty.

In the second experiment rats were trained either on a brightness or spatial discrimination reversal problem in the Ross-maze. It was found that overtraining facilitated reversal performance in the spatial task but not in the brightness problem. The theoretical implications of these results were discussed.  相似文献

The first purpose of this study was to investigate whether lesions in the temporal region may affect acquisition or retention of a discrimination task. In Experiment 1, rats with lesions of the temporal cortex (TC), the lateral entorhinal cortex (LEC), or their interconnections were tested postoperatively in simultaneous brightness discrimination. The results show that neither TC lesions nor LEC lesions affected acquisition of the task, and only LEC lesions impaired retention. TC/LEC transections impaired both acquisition and retention. The second purpose was to investigate effects of hippocampal lesions and perforant path transections on the discrimination task (Experiment 2). Both hippocampal and perforant path lesions impaired acquisition of the task, whereas retention was unaffected. It is suggested that TC and LEC are primarily involved in information storing and that hippocampal function is primarily involved in information processing.  相似文献

In a recent study we established that following a partial acquisition of a brightness discrimination escape task (15 trials), Sprague-Dawley albino rats exhibited a long-term spontaneous improvement of performance (LTSI) after 7 to 14 days. Some evidence suggests a relation between the strength of the initial training and the delay of the optimal retention performance. This study investigates such a possibility in studying performance of rats (number of trials needed to reach a criterion of 10 errorless trials) following nine different retention intervals: 10 min, and 1, 3, 5, 7, 10, 14, 21, and 28 days, after a strengthened initial training (25 trials). Under those conditions, LTSI occurred after a delay of 5 to 7 days. Theoretical implications for an inverse relationship between the strength of initial training and the length of delay leading to LTSI are discussed.  相似文献

A discrimination box containing two levers with a light above each was used to train eight rats to press beneath the brighter light for a milk reinforcer. The brighter light was held constant and the comparison light was varied to produce 12 brightness differences. The animals were run under two experimental methods: the block method in which each brightness level comparison was presented for a block of 11 contiguous trials, and the staircase method in which the sequence of brightness comparisons was determined by the correctness of the response on the preceding comparison. The block method produced a smaller differential brightness threshold and a larger change in discrimination performance for stimulus magnitude changes than did the staircase method.  相似文献

Rats receiving polyunsaturated fatty acid (PUFA) deficient diets during the perinatal period showed in adult age undisturbed acquisition of a footshock motivated brightness discrimination task, but a significant impairment of retention. The same effects on retention were obtained in rats receiving the PUFA deficient diet in adulthood, when the behavioral parameters were investigated at the end of the dietary treatment.  相似文献

This series of experiments investigates possible relations between increases in paradoxical sleep (PS), persisting for several days after an avoidance training, and improvement of retention performance that occurred 3 days following partial training in a brightness discrimination Y-maze shock-avoidance task. Sprague-Dawley rats were trained in the Y-maze and PS deprived for 24 h either immediately or 24, 48, or 72 h following initial training. Contrary to what was expected, the results indicated that PSD immediately following the training session enhanced the avoidance performance after a 7-day retention interval. PSD at later times had no effect. Experiment 2 indicated that this effect was obtained only for PS-deprived animals and not for those placed in the PSD situation, but on larger platforms. Thus enhancement of the avoidance performance was not due to increases in stress or arousal caused by PSD-associated factors. Experiment 3 showed that the facilitative effect of a non-delayed 24-h PSD was obtained immediately thereafter as well as 24 h later, demonstrating that this effect was not due to any PS rebound which might have occurred following the PSD. Alternative explanations for these unexpected results are discussed.  相似文献

Pre-test exposure to training-related cues is known to improve subsequent retention performance. To identify brain regions engaged in processes promoted by retrieval cues, a brain imaging approach using the [6-14C]glucose autoradiographic technique was used. Sprague-Dawley rats trained in a brightness discrimination avoidance task were submitted to different cueing conditions after a 1- or a 21-day training-to-test interval (TTI). Animals were either non-cued, cued with a box, or cued with a box and the light that served as a discriminative stimulus. Effects of the different cueing conditions on retention performance or on metabolic activity in 58 different brain regions were investigated. Rats cued with the light exhibited a subsequent improvement of their retention performance relative to controls, when tested at the 1- but not 21-days TTI, confirming our previous results. At the 1-day retention interval, a comparison between rats cued with the box and rats cued with the box and the light showed that the light cue significantly increased glucose uptake in a neuronal network composed of the lateral, basal, and central nuclei of the amygdala, the anterior and suprachiasmatic hypothalamic nuclei, the nucleus accumbens, the medial septum, and the insular cortex. In contrast, at the 21-day retention interval, both groups demonstrated similar cerebral metabolic activity. The present results indicate that exposure to a light cue increased metabolic activity in the previously mentioned brain structures only when the light acted as an effective retrieval cue, suggesting an involvement of this network in the processes triggered by a retrieval cue. Arguments are provided supporting the notion that the amygdala may play a key role in these processes. Whether the amygdala is a part of a neural network involved in retrieval processes or in neuromodulating systems that favour the efficacy of retrieval processes is also discussed.  相似文献

Results from previous studies suggest that the entorhinal cortex may be involved in mnemonic processes. The present study was carried out to investigate whether disruption of fibre connections between the temporal cortex and lateral entorhinal area may impair retention of a pre-operatively acquired simultaneous brightness discrimination task. The lesion resulted in a severe impairment in retaining the discrimination task (Experiment 1). The retention deficit could not be traced into the hippocampal formation by making perforant path lesions or hippocampal lesions (Experiment 2). The results indicate that the lateral entorhinal cortex is more crucial for reference memory than the hippocampal formation.  相似文献

The aim of this investigation was to determine what effect variation in the conditions of cue presentation and response availability has on the visual discrimination performance of monkeys with bilateral inferior temporal ablations. Six animals were trained on one of two versions of a test of successive brightness discrimination. Common to both versions of this test was the availability of a single lever, manipulation of which in the presence of the positive cue constituted the correct response. Performance on this test was compared with performance on a pattern discrimination, for which the customary simultaneous two-choice procedure was used. It was found that the temporal removals were followed by definite impairment on the pattern discrimination (as expected), but no consistent change in efficiency at brightness discrimination could be attributed to the lesions. Alternative interpretations, making reference to the amount of pre-operative training on the brightness discrimination or the lack of differentiation between the correct and alternative responses in this test, are discussed.  相似文献

The selective attention and stimulus generalization interpretations of the easy-to-hard effect were tested by training one group of rats on an easy brightness discrimination and “reversing” them on a more difficult brightness discrimination. A control group, initially trained on the difficult discrimination, was reversed at the same time as the experimental group. The experimental group learned the reversal more rapidly and this was interpreted as supporting the selective attention interpretation.  相似文献

The impact of an acute circadian disruption on learning and memory in male and female rats was examined. Circadian disruption was elicited using a brief series of photoperiod shifts. Previous research using male rats showed that acute circadian disruption during acquisition of a spatial navigation task impaired long-term retention and that chronic circadian disruption impaired acquisition of the same task. However, the long-term effects of acute circadian disruption following circadian re-entrainment and whether sex differences in response to circadian disruption exist are still unknown. For the present study, rats were trained on the standard, spatial version of the Morris water task (MWT) and a visual discrimination task developed for the eight-arm radial maze. After reaching asymptotic performance, behavioural training was terminated and the experimental group experienced a series of photoperiod shifts followed by circadian re-entrainment. Following circadian re-entrainment, the subjects were given retention tests on the MWT and visual discrimination task. Following retention testing, an extra-dimensional shift using the eight-arm radial maze was also performed. An acute episode of circadian disruption elicited via photoperiod shifts negatively impacted retention of spatial memory in male and female rats. Retention of the visual discrimination task and the ability to detect extra-dimensional shifts were not impaired. The observed impairments on the MWT indicate that hippocampal representations are susceptible to a small number of photoperiod shifts even if the association is acquired prior to rhythm manipulation and retention is assessed following rhythm stabilization. Effects were limited to a hippocampus-dependent task, indicating that impairments are specific, not global.  相似文献

The performance of rats over 12 brightness discrimination reversals was studied under two experimental conditions. Under one condition all visual cues external to the apparatus were eliminated so that only the relative positions of the discriminanda could serve as a visual cue to spatial position. Under the other condition all visual cues to position were eliminated. Under the former condition performance deteriorated with successive reversals but under the latter condition performance improved. Implications of these results for theories of successive reversal improvement were discussed and two possible explanations were suggested.  相似文献

Fetal brain tissue from the occipital or the frontal cortex was implanted into the damaged occipital cortex of adult rats. The animals receiving grafts of embryonic frontal cortex showed partial restoration of brightness discrimination while recipients given homologous implants of occipital cortex were as impaired as those animals with lesions alone. Neither frontal nor occipital grafts aided in the performance of a pattern discrimination problem; both groups of brain-damaged animals were unable to learn the task. Nonetheless, both groups of animals had viable and enlarged grafts with similar neuronal and glial profiles.  相似文献

Previously, we reported that posttraining paradoxical sleep deprivation (PSD) resulted in an enhancement of the subsequent avoidance performance for rats trained for 15 trials in a Y-maze brightness avoidance discrimination task. A series of experiments were conducted to try to further understand the reasons for results which were contrary to those of the bulk of the sleep-learning literature. Experiment 1 investigated the effectiveness of the PSD technique. Rats (N= 4) were sleep recorded while residing on a “swimming pool” apparatus for 24 h. Compared to their baseline values, all animals showed a very large reduction in paradoxical sleep and spent significantly more time awake. Slow-wave sleep was unchanged. In Experiment 2, proactive motor effects were tested. Rats were deprived of PS for 24 h and then tested in a hole board motor activity task. There was a slight effect of PS deprivation on the day following the PSD and no effect when the rats were retested 1 week later. Experiment 3 investigated possible proactive effects of PSD on avoidance performance. Rats exposed to PSD in the 24 h before training in the Y-maze task did not demonstrate any facilitative effect on the subsequent avoidance performance. Experiment 4 investigated the possibility that the PSD facilitative effect could be due to partial training. Rats were given 75 acquisition trials in the brightness discrimination Y-maze avoidance before being subjected to 24 h of PSD. PS-deprived animals showed superior avoidance scores compared to non-PSD controls when retested 24 h later. In Experiment 5, the same strain of rats (N= 11) were sleep recorded after exposure to a partial acquisition in a Y-maze brightness avoidance discrimination task. They were then continuously monitored for 4 consecutive days. The percent PS for the Trained rats was significantly lower than that for the Control animals. This drop in percent PS was not confined to any particular time period in the 24-h day. None of the other sleep parameters reached significance. Analyses of the present results suggest that PSD exerts its facilitative effects on posttraining consolidation processes. We present arguments suggesting that PSD can have effects opposite to those generally reported, in animals demonstrating poor avoidance abilities, in an avoidance task.  相似文献

Several experiments have found complete retention of conditioned suppression when overall suppression to the signal for shock was measured. The present experiment examined retention of conditioned suppression but did so with a paradigm that produced temporal discrimination of shock occurrence. Nine rats were exposed to a flashing light signal of 5-min duration that always terminated with a shock. After several months, a temporal discrimination was well established, as shown by maximum suppression toward the end of the signal period. After remaining in the home cage for 25 days, the rats were again subjected to the conditioning procedure. The overall level of suppression remained the same but the temporal discrimination was not observed.  相似文献

The present experiments determined the consequences of blocking muscarinic cholinergic receptors of the prelimbic (PL) cortex in the acquisition and retention of an odor-reward associative task. Rats underwent a training test (five trials) and a 24-h retention test (two retention trials and two relearning trials). In the first experiment, rats were bilaterally infused with scopolamine (20 or 5 microg/site) prior to training. Although scopolamine rats showed acquisition equivalent to PBS-injected controls, they exhibited weakened performance in the 24-h retention test measured by number of errors. In the second experiment, rats were injected with scopolamine (20 microg/site) immediately or 1 h after training and tested 24 h later. Scopolamine rats injected immediately showed severe amnesia detected in two performance measures (errors and latencies), demonstrating deficits in retention and relearning, whereas those injected 1 h later showed good 24-h test performance, similar to controls. These results suggest that muscarinic transmission in the PL cortex is essential for early memory formation, but not for acquisition, of a rapidly learned odor discrimination task. Findings corroborate the role of acetylcholine in consolidation processes and the participation of muscarinic receptors in olfactory associative tasks.  相似文献

Effects of vasopressin were measured in a nonspatial working memory task: food-reinforced, delayed matching to sample. Subcutaneous injections of 0.2 microgram of lysine vasopressin (LVP) or saline were alternately administered to Sprague-Dawley rats after the presentation of the sample and compared to the effect of the same treatments given to a yoked control group of rats. Different durations of sample presentation (5 and 30 min) and various retention intervals (10 min, 3 h, 24 h) were selected. The results showed that LVP never facilitated retention performance: there was no improvement under conditions of weak memory (short presentation of the sample and long retention interval); moreover LVP abolished the facilitation normally obtained when either the length of the sample presentation was longer or the duration of the retention interval was shorter. The performance after the injection of the peptide was differentially impaired, according to brightness of the sample which had been presented: after the presentation of the white box, LVP injections lead to more errors and after that of the black one the treatment induced an increase in latencies of response. All these data may suggest that the physiological consequences of hormonal modifications triggered by the LVP injection are processed along with the stimuli of the learning episode and interfere with the learned positive value of the sample.  相似文献

This study examined the effects of a dopamine D1 antagonist, SCH23390, infused into the prelimbic–infralimbic areas on the acquisition of a response and visual-cue discrimination task, as well as a shift from a response to a visual-cue discrimination and vice versa. Each test was carried out in a cross-maze. The response discrimination required learning to always turn in the same direction (right or left) for a cereal reinforcement. The visual-cue discrimination required learning to always enter the arm with the visual cue. In experiment 1, rats were tested on the response discrimination task, followed by the visual-cue discrimination task. In experiment 2, the testing order was reversed. Bilateral infusions of SCH23390 (0.1 or 1 μg/0.5 μL) into the prelimbic–infralimbic areas did not impair acquisition of the response or visual-cue discrimination tasks. SCH23390 injections at 1 μg, but not 0.1 μg impaired performance when shifting from a response to a visual-cue discrimination, and vice versa. Analysis of the errors revealed that the deficit was due to perseveration of the previously learned strategy. These results suggest that activation of dopamine D1 receptors in the prelimbic–infralimbic areas may be critical for the suppression of a previously relevant strategy and/or generating new strategies.  相似文献

Retrograde and anterograde amnesic effects of excitotoxic lesions of the rat retrosplenial cortex (RS) and hippocampus (HPC) were investigated. To test retrograde amnesia, rats were trained with two-arm place discrimination in a radial maze 4 wk and 1 d before surgery with a different arm pair, respectively. In the retention test 1 wk after surgery, both lesion groups showed temporally ungraded retrograde amnesia. To test anterograde amnesia, animals were trained after surgery to discriminate three arm pairs successively within a day, and then after interposition of 1- to 4-wk intervals, one of these pairs, respectively, was tested for retention. RS-lesioned rats could acquire these pairs of place discriminations rapidly but showed a retention interval-dependent impairment in the retention test. Conversely, HPC-lesioned rats took more sessions to acquire these pairs than did the control group, and their retention was approximately 70% of correct performance regardless of retention interval. Results suggest that RS and HPC have different roles in spatial memory and that RS is important for remote memory process.  相似文献
