Involvement of BDNF receptor TrkB in spatial memory formation   总被引:11,自引:0,他引:11       下载免费PDF全文
The N-methyl-D-aspartate (NMDA) receptors are involved in long-term potentiation (LTP), and are phosphorylated by several tyrosine kinases including a Src-family tyrosine kinase Fyn. Brain-derived neurotrophic factor (BDNF) is a neurotrophin, which also enhances hippocampal synaptic transmission and efficacy by increasing NMDA receptor activity. Here, we show that Fyn is a key molecule linking the BDNF receptor TrkB with NMDA receptors, which play an important role in spatial memory formation in a radial arm maze. Spatial learning induced phosphorylation of TrkB, Fyn, and NR2B, but not NR2A, in the hippocampus. Fyn was coimmunoprecipitated with TrkB and NR2B, and this association was increased in well-trained rats compared with control animals. Continuous intracerebroventricular infusion of PP2, a tyrosine kinase inhibitor, in rats delayed memory acquisition in the radial arm maze, but PP2-treated animals reached the same level of learning as the controls. The phosphorylation of Fyn and NR2B, but not TrkB, was diminished by PP2 treatment. Our findings suggest the importance of interaction between BDNF/TrkB signaling and NMDA receptors for spatial memory in the hippocampus.  相似文献

Three experiments investigated how the frequency of exposure to particular exemplars influenced 10-month-old infants' differentiation of land and sea animals in an object-examining task. In Experiments 1 and 2, one category exemplar was presented more frequently than the others during familiarization (i.e., that exemplar was presented on 6 of 12 familiarization trials, and 3 other exemplars were each presented on 2 familiarization trials). For half of the infants, the frequent exemplar was similar to other category exemplars (e.g., a zebra if the familiarization category was land animals), and for half the frequent exemplar was not similar to many other category exemplars (e.g., a rabbit). Infants who frequently experienced the similar exemplar formed an exclusive category, and differentiated land and sea animals. Infants who frequently experienced a dissimilar exemplar, in contrast, formed an inclusive category, and failed to differentiate between land and sea animals. In Experiment 3, infants received frequent experience with a set of similar or dissimilar exemplars, and the same pattern was observed. Thus, 10-month-old infants are sensitive to the distribution of the exemplars to which they are exposed, and they form different category boundaries depending on that distribution.  相似文献

In a series of experiments, a visual search task was used to test the idea that biologically relevant threatening stimuli might be recognized very quickly or capture visuo-spatial attention. In Experiment 1, there was evidence for both faster detection and faster search rates for threatening animals than for plants. However, examination of the basis of this effect in Experiment 2 showed that it was not due to threat per se, as detection and search rate advantages were found for pleasant rather than threatening animals compared to plants. In Experiment 3, participants searched for the plants and pleasant and threatening animals used in Experiments 1 and 2, among a fixed heterogeneous selection of non-target items. There was no search rate or detection advantage for threatening animals compared to pleasant animals or plants. The same targets and non-targets as those used in Experiment 3 were also used in Experiment 4. In Experiment 4, participants searched for targets that were presented either close to or distant from an initial fixation point. There was no evidence for a "threat" detection advantage either close to or distant from the cross. Finally, an experiment was conducted in which target categories (fruit, flowers, and animals) were not pre-specified prior to each trial block. There were no differences in reaction times to detect pleasant animals, threatening animals, or fruit. We conclude that the visual search paradigm does not readily reveal any biases that might exist for threatening stimuli in the general population.  相似文献

Water-deprived rats in a pair competing for a single source of water quickly establish a firm relationship during which one rat drinks consistently more (dominant) than the other (submissive) animal. This relationship is formed during the first competition and is very stable during subsequent tests. Exposure of dominant rats to a severe (18 hrs immobilization), but not a mild (2 hrs immobilization), stressor reduced markedly aggressive behavior and inverted transiently the dominant submissive relationship of the pairs. Exposure of submissive rats to the severe stressor resulted in only minor reductions of aggressive behavior in these animals. Prestress anxiety predicted stress effects in the dominant animals in that high-anxious animals lost more dominant behavior and weight during stress as compared with low-anxious rats. Thus, severe stress can transiently reduce dominant but not submissive behavior during water competition and high-anxious rats are more prone to lose their aggressive behavior.  相似文献

Chicks learned to find food hidden under sawdust by ground-scratching in the central position of the floor of a closed arena. When tested inan arena of identical shape but a larger area, chicks searched at 2 different locations, one corresponding to the correct distance (i.e., center) in the smaller (training) arena and the other to the actual center of the test arena. When tested in an arena of the same shape but a smaller area, chicks searched in the center of it. These results suggest that chicks are able to encode information on the absolute and relative distance of the food from the walls of the arena. After training in the presence of a landmark located at the center of the arena, animals searched at the center even after the removal of the landmark. Marked changes in the height of the walls of the arena produced some displacement in searching behavior, suggesting that chicks used the angular size of the walls to estimate distances.  相似文献

Although research has shown that media coverage of traumatic events can evoke intense fear in spectators, few studies have examined the mechanisms underlying this effect. Drawing from appraisal theories, this article investigates whether viewers' fear responses to traumatic reports are mediated by the appraisal of personal relevance, as well as whether positive framing of news stories is capable of dampening fear. In a 2 × 2 between-subjects design, participants watched one of two reports about mutilations caused by either an event with high signal potential (bacterial infection) or an event with low signal potential (explosion of land mines). Each of the reports could be either positively or negatively framed. Participants were asked to report their emotions in response to the news story and to answer questions concerning their cognitive appraisal. Results showed that the report with mutilations caused by bacterial infection elicited more fear than the report with mutilations caused by land mine explosions. This effect was mediated by the dimensions of suddenness, unpleasantness, personal relevance, and coping potential. Positively framed reports generated less fear than negatively framed reports, and this effect was mediated by the dimensions of coping potential and unpleasantness. The implications for media research are discussed.  相似文献

A recent study (Tommasi & Thinus-Blanc, 2004) found that when rats trained to search for food hidden in the center of a square-shaped enclosure were tested in the absence of food, they searched precisely in the center of the enclosure. Transfer tests carried out in enclosures differing in shape or size (rectangular-, large square-, and equilateral triangle-shaped) showed that rats searched in the geometric center of the novel environments. In the present work, half of those rats were lesioned bilaterally to the hippocampus and were then re-trained and re-tested in the same task, to be compared to sham rats that received the same retraining/re-test protocol. Results show that lesioned rats, although unimpaired in relearning to localize the center of the square-shaped enclosure, performed considerably worse than sham rats in the transfer tests in the enclosures differing in shape, suggesting a relevant role of the hippocampus in geometry-driven place generalization.  相似文献

The aim of the present study was to evaluate the occurrence of fear sensitization in rats previously treated with an inhibitor of the NO syntheses and submitted to Trial1/Trial2 plus-maze (PM) procedure. Male Wistar rats received a systemic treatment with N(omega)-nitro-L-Arginine-methyl ester (L-NAME; 5, 10 or 50 mg kg(-1)) and were submitted to PM Trial1. In the following day the animals were re-exposed to the PM with no drug administration (Trial2). Some standards spatial-temporal measures, such as the percentage of entries (% Open arm entries) and time spent (% Open arm time) in the open arms and risk assessment frequency were recorded and used to estimate the animal level of fear sensitization in PM Trial2. The results showed that animals receiving L-NAME (50 mg kg(-1)) displayed increased % Open arm entries and % Open arm time in Trial2 in relation to the group receiving 0.9% saline, which is compatible with impaired fear/anxiety acquisition during Trial1/Trial2 PM procedure. In addition, rats treated with L-NAME (50 mg kg(-1)) exhibited low level of risk assessment in Trial2 in relation to the group treated with 0.9% saline, which indicates low level of fear/anxiety during PM re-exposure. The number of entries into the enclosed arms was not changed by any L-NAME treatment, which suggests no bias of the drug treatments on animal locomotor activity. The data suggest that NO synthesis may mediate the fear sensitization process in the PM.  相似文献

Twenty-four-month-old and 4-month-old rats were trained on a peak-interval procedure, where the time of reinforcement was varied twice between 20 and 40 sec. Peak times from the old rats were consistently longer than the reinforcement time, whereas those from younger animals tracked the 20- and 40-sec durations more closely. Different measures of performance suggested that the old rats were either (1) systematically misremembering the time of reinforcement or (2) using an internal clock with a substantially greater latency to start and stop timing than the younger animals. Old rats also adjusted more slowly to the first transition from 20 to 40 sec than did the younger ones, but not to later transitions. Correlations between measures derived from within-trial patterns of responding conformed in general to detailed predictions derived from scalar expectancy theory. However, some correlation values more closely resembled those derived from a study of peak-interval performance in humans and a theoretical model developed by Cheng and Westwood (1993), than those obtained in previous work with animals, for reasons that are at present unclear.  相似文献

In two separate experiments, rats with bilateral lesions of the superior colliculus showed significantly poorer relearning of a horizontal/vertical stripe discrimination than control animals. In Experiment 1, all animals showed disruption of performance when a stimulus--response (S--R) separation was introduced by raising the stimuli above the site of responding. However, the colliculectomized rats were much more disturbed by the S--R separation than were animals in the control group. In Experiment 2, all animals showed lower performance levels when conflicting patterns were introduced into the upper portion of the stimulus doors, but this time the rats with collicular lesions were less disturbed than the control animals. It is suggested (a) that when the stimulus and response sites are discontiguous, animals must make an appropriate orienting response in order to effectively sample the visual stimuli and (b) that lesions of the superior colliculus alter performance by interfering with this orienting behavior. The impairment in relearning is tentatively attributed to the absence of preoperative overtraining on the discrimination task.  相似文献

The immediate-early gene hzf-3, also known as nurr1, is a member of the inducible orphan nuclear receptor family and is one candidate in the search for genes associated with learning and memory processes. Here we report that acquisition of a spatial food search task is accompanied by elevated levels of hzf-3 mRNA in the hippocampus. Adult male Long-Evans rats were handled, food-restricted, and allowed to habituate to the maze prior to training. During acquisition, rats were given one training session per day for 5 days. Each training session consisted of five trials in which animals searched the maze for food located in 4 of 16 holes in the floor of the maze. Training resulted in spatial acquisition of the task. Northern blot analysis showed significant increases in hippocampal hzf-3 mRNA 3 h after training in the maze. Next, brains were obtained from Naive, Habituated, Day 1, Day 3, and Day 5 animals and processed for in situ hybridization. The results showed significant increases of hzf-3 mRNA in CA1 and CA3 subregions of the dorsal hippocampus during acquisition of the task. We conclude that expression of the hzf-3 gene in the brain is associated with long-term spatial memory processes. The present results are the first to implicate an orphan nuclear receptor in long-term information storage in the hippocampus.  相似文献

Syracuse high- and low-avoidance Long-Evans rats (Rattus norvegicus; SHA/Bru and SLA/Bru) were selectively bred for good and poor active-avoidance learning. However, SLA/Bru animals are superior to SHA/Bru rats in conditioned suppression and passive avoidance learning. In this experiment, saccharin taste and almond odor were the components of a compound conditioned stimulus (flavor) in an illness-induced aversive conditioning paradigm. SLA/Bru rats (n = 17) showed stronger conditioned flavor, taste, and odor aversion than did SHA/Bru animals (n = 18). Unselected Long-Evans rats (n = 18) were intermediate between the selected strains. SLA/Bru and Long-Evans rats showed taste-potentiated odor aversions in this experiment, whereas SHA/Bru animals did not. The results provide evidence that genetic factors, as exemplified by the different strains, are importantly involved in the mechanisms underlying interoceptive and exteroceptive aversive conditioning.  相似文献

KCl water preference was examined in diet-replete, sodium-and sodium—potassium-deficient postpubertal rats of different ages. While diet-replete rats displayed a KCl preference-aversion profile similar to that found using other salts, when retested 30 days later, these same animals had no KCl preference. Na-and Na—K-deficient rats preferred KCl over a broad range of concentrations when tested in early postpuberty and the Na—K-deficient group had a markedly reduced KCl preference threshold (0.12%) when compared to all other groups in the study. But Na-and Na—K-deficient rats older by 2 1/2 months did not prefer KCl. The notion of specific food hungers can be invoked to account for some of the preferences. But hedonics, age and KCl experience must also be considered based on (1) the quantities of KCl consumed, (2) the preference reversals within animals, as well as (3) the significantly increased heart—and kidney—body weight ratios in the rats with the greatest KCl preference.  相似文献

Psychotherapy has a rich past and great potential. However, psychotherapists should be aware of several areas of strength as well as some problem areas. Strengths include the benefits of a strong therapeutic alliance, a diverse range of theories, and quality research that documents the effectiveness of psychotherapy. Weaknesses include conflicting theories, financial matters, and misunderstandings between clinicians and researchers. Hopefully, mental health professionals can focus on mining the gold from the gold mines while also working to understand and disarm the land mines.  相似文献

Recent research has examined whether animals possess metacognition, or the ability to monitor their knowledge states. However, the extent to which animals actively control their knowledge states is still not well delineated. Although organisms might be capable of seeking information when it is lacking, it does not mean that it is always adaptive to do so. In the present set of experiments, we examined the flexibility of this behavior in captive orangutans (Pongo abelii; two adults and one juvenile) in a foraging task, by varying the necessity of information-seeking, the cost associated with it, the likelihood of error, and the value of the reward. In Experiment 1, subjects searched for information most often when it was "cheapest" energetically. In Experiment 2, subjects searched for information most often when the odds of making an error were the greatest. In Experiment 3, subjects searched for information more when the reward was doubled in value. In Experiment 4, adult subjects adapted to risk/benefit trade-offs in their searching behavior. In every experiment, subjects sought information more often when they needed it than when they already knew the solution to the problem. Therefore, the current research suggests that information-seeking behavior in orangutans shows a sophisticated level of flexibility, comparable to that seen in human children, as they appear to "play the odds" when making the decision to seek information or not.  相似文献

The detection and subsequent removal of land mines and unexploded ordnance (UXO) from many developing countries are slow, expensive, and dangerous tasks, but have the potential to improve the well-being of millions of people. Consequently, those involved with humanitarian mine and UXO clearance are actively searching for new and more efficient detection technologies. Remote explosive scent tracing (REST) using trained dogs has the potential to be one such technology. However, details regarding how best to train, test, and deploy dogs in this role have never been made publicly available. This article describes how the key characteristics of applied behavior analysis, as described by Baer, Wolf and Risley (1968, 1987), served as important objectives for the research and development of the behavioral technology component of REST while the author worked in humanitarian demining.  相似文献

Handled (Day 1-22) and non-handled infantile Wistar rats were tested in maturity for the partial reinforcement extinction effect (PREE) and the partial punishment effect (PPE). In Experiments 1 and 2, mature male and female rats were trained to run in an alley for food reward on a 1-trial/day schedule. In the PREE paradigm (Experiment 1), the partially reinforced group (PRF) received reinforcement on a quasi-random 50% schedule, while the continuously reinforced group (CRF) received reinforcement on every trial. In the test stage, both groups were given extinction training. In the PPE paradigm (Experiment 2), the partially punished (PP) group received, together with continuous reinforcement, shocks on a quasirandom 50% schedule, while the continuously reinforced group was reinforced on every trial. In test, all animals were given both reinforcement and shock on every trial. In Experiment 1, PREE—i.e. increased resistance to extinction in the PRF as compared to the CRF group—was more pronounced in the handled animals. More specifically, no PREE was obtained in the non-handled males, and in the non-handled females the PREE was reduced compared to the handled females. The results of Experiment 2 revealed no effect of handling or sex on PPE, that is, increased resistance to punishment in the PP group as compared to the CRF group was evident in all four conditions. In Experiment 3, handled and non-handled male rats were tested for the PREE using a multi-trial procedure in an operant chamber. PREE was obtained in the handled but not in the non-handled animals. The implications of these results for the differential effects of handling on male and female rats and the distinction between the PREE and PPE paradigms are discussed.  相似文献

The effect of visual distracting stimuli upon the straight alleyway performance of dorsal hippocampectomized Wistar rats was investigated. In comparison with control animals it was observed that dorsal hippocampectomized animals (1) ambulated more during the preexposure phase, (2) acquired at the same rate a running response for food (training phase), (3) reacted similarly to a new visual stimulus (black cards) presented in a sector of the alleyway, and (4) habituated to successive presentations of that stimulus in the same place. (5) However, dorsal hippocampectomized rats did not react, unlike the controls, to the presentation of the same stimulus in another place of the alleyway but (6) reacted to the visual pattern change of the stimulus (now black/white check cards) in the same place. These results indicate that under certain experimental conditions, hippocampus-lesioned animals are capable of interrupting a running response for food in order to explore a new conspicuously located stimulus, habituate to repeated presentations of that stimulus, and to react to a new pattern of visual stimulation. They suggest that hippocampectomized rats do not lose the capacity to react to a new stimulus; the disruption seems to be related to the spatial context of stimulus presentation, supporting a spatial mapping hypothesis of hippocampal function.  相似文献

Three experiments examined how a flavour modified the development of aversions to the place in which it was presented and paired with a reinforcer. Experiment 1 confirmed the cue-to-consequence effect: rats made sick after exposure to a flavour in a novel place (Skinner boxes) displayed stronger aversions than shocked animals when they were presented with the flavour in a second, familiar place; rats shocked after exposure to water acquired stronger place aversions than animals made sick. Experiment 2 confirmed the potentiation effect: rats made sick after exposure to the flavour developed stronger place aversions than those made sick after ingestion of water. This experiment also revealed that rats shocked after exposure to the flavour likewise acquired stronger place aversions than the animals shocked after ingestion of water. Experiment 3 replicated this flavour potentiation of place aversions based on shock. The results are not consistent with Garcia, Lasiter, Bermudez-Rottoni, & Deems' (1985) account of how a flavour interacts with an aversive reinforcer to modify what is learned about its associates in a compound.  相似文献

The present study examined the influence of short- and long-term chronic intermittent immobilization stress throughout the brain and on the adrenal medulla of intact rats using Fos-like immunoreactivity (Fos-LI) as a marker of cellular activation. The effect of adreno-medullectomy on the central nervous system (CNS) response to chronic immobilization stress was also examined. It was found that control unoperated, unstressed rats had no Fos-LI cells in the brain or in the adrenal medulla. In intact rats, neither short term (1 week) nor long term (4 weeks) chronic intermittent immobilization stress produced significant increases in Fos-LI in the CNS compared with control animals. However, marked increase in the number of Fos-LI cells was observed in the adrenal medulla of animals stressed for 4 weeks compared with control, unstressed animals or those stressed for 1 or 2 weeks. In adreno-medullectomised rats, 4 weeks, but not 1 week, chronic immobilization stress produced significant increases in numbers of Fos-LI neurons in the paraventricular hypothalamic and supraoptic nuclei and the medial amygdala compared with intact animals stressed for a similar period of time. It is concluded that long term stress produces chronic Fos-LI in the adrenal medulla and that adreno-medullectomy increases the Fos response of the PVN, supraoptic nucleus and medial amygdala to long term stress.  相似文献
