In rodents, fear conditioned responses are more pronounced toward olfactory stimulus, since olfaction is a dominant sense in these subjects. The present study was outlined to investigate if the association between coffee odor (CS1) and electrical footshock (US) would be an effective model for the study of fear-induced behavior and whether compounds used in humans for emotional-related disorders such as midazolam, propranolol, or scopolamine, applied during the different stages of fear conditioning (acquisition, consolidation and expression), affect the defensive responses to both, the olfactory CS1, and the context (CS2) where the CS1 had been presented (second order conditioning). The results revealed that five pairings between coffee odor (CS1) and electrical footshock (US) were able to elicit consistent defensive responses and a second order conditioning to the context (CS2). Midazolam (0.375–0.5 mg/kg; i.p.) treatment was able to interfere with the CS1–US association and with the consolidation of the aversive information. The propranolol (5–10 mg/kg; i.p.) treatment interfered with the CS1–US association, with the retention of fear memory and with the CS1–CS2 association. Propranolol also attenuated the expression of conditioned fear responses when applied before the CS1 test session. Scopolamine (0.6–1.2 mg/kg; i.p.) treatment impaired the acquisition of CS1–US and CS1–CS2 associations, and also disrupted the expression of conditioned fear responses when injected prior to the CS1 test session. These findings have pointed out the usefulness for the olfactory fear conditioning paradigm to investigate drug effects on the acquisition, consolidation and expression of fear conditioned responses.  相似文献

Protein-deprived rats were given, on one day, a balanced mixture of amino acids followed by access to protein-free food having a distinctive odour. On another day, an imbalanced (histidine-free) amino acid mixture was given just before food having another odour. The rats afterwards preferred the balance-paired odour to the imbalance-paired odour. The preference was acquired whether the duration of odour presentation, or the amount of odourised food taken, was kept constant on the two conditioning days. Retention of the preference seemed unattenuated after 4 weeks. An attraction to the balance-paired odour (relative to odours paired with a water load) contributed to the acquired preference. There was also a relative aversion to unfamiliar odours when they had been paired with imbalance. Such acquired chemosensory control of preferences, together with an anorexigenic effect of imbalanced amino acid mixtures, can account for characteristics of feeding behaviour under conditions in which the diet is deficient in an essential amino acid.  相似文献

Thirty-two Ss between 22 and 59 years of age yielded detection thresholds for 4 odorants over 4 sessions. The thresholds decreased and reliability increased over the course of testing. High intercorrelations between odorants and the stability of an S's relative position within the threshold distributions showed that a general factor of sensitivity dominated the outcome. Age contributed strongly to intersubject variation. Even among these nonelderly individuals, it accounted for up to 2 orders of magnitude in threshold performance. Other important factors included superiority of the right nostril and a negative correlation between the mean and variance of threshold distributions. Scant attention to the correlation may have contributed to overestimation of the frequency and specificity of specific anosmia. A clinically relevant outcome was that measurement of threshold for diagnostic purposes can generally rely on just 1 odorant.  相似文献

The widely used Pavlovian fear-conditioning paradigms used for studying the neurobiology of learning and memory have mainly used auditory cues as conditioned stimuli (CS). The present work assessed the neural network involved in olfactory fear conditioning, using olfactory bulb stimulation-induced field potential signal (EFP) as a marker of plasticity in the olfactory pathway. Training consisted of a single training session including six pairings of an odor CS with a mild foot-shock unconditioned stimulus (US). Twenty-four hours later, the animals were tested for retention of the CS as assessed by the amount of freezing exhibited in the presence of the learned odor. Behavioral data showed that trained animals exhibited a significantly higher level of freezing in response to the CS than control animals. In the same animals, EFPs were recorded in parallel in the anterior piriform cortex (aPC), posterior piriform cortex (pPC), cortical nucleus of the amygdala (CoA), and basolateral nucleus of the amygdala (BLA) following electrical stimulation of the olfactory bulb. Specifically, EFPs recorded before (baseline) and after (during the retention test) training revealed that trained animals exhibited a lasting increase (present before and during presentation of the CS) in EFP amplitude in CoA, which is the first amygdaloid target of olfactory information. In addition, a transient increase was observed in pPC and BLA during presentation of the CS. These data indicate that the olfactory and auditory fear-conditioning neural networks have both similarities and differences, and suggest that the fear-related behaviors in each sensory system may have at least some distinct characteristics.  相似文献

Five experiments investigated the phenomenon that attitude formation is not confined to the co-occurrence of an attitudinal object with an evaluated experience. The pairing of a target with a (dis)liked person not only affects the evaluation of the previously neutral person but spreads to other individuals who are (pre)associated with the target (spreading attitude effect). Experiments 1 and 2 provided evidence for the spreading attitude effect in appetitive as well as aversive evaluative conditioning. Experiment 3 showed that the spreading attitude effect is a robust phenomenon resistant to extinction. Experiment 4 demonstrated that attitude spread can be transferred to 2nd-order conditioning. Finally, Experiment 5 supports the notion that the spreading attitude effect is not dependent on cognitive resources. Implications for social as well as applied psychology are discussed.  相似文献

Sucrose was used to reinforce heart rate (HR) increases in one group of unrestrained rats and to reinforce decreases in another. A third group received noncontingent sucrose presentations, and a fourth was presented with an empty dipper. After 10 conditioning sessions HR, as measured by changes in tonic levels, was significantly greater in the Fast group than in the Noncontingent group, which in turn maintained significantly higher levels than the Slow group. There was no difference between the Noncontingent and No-sucrose groups. Behavioral observations indicated that the Fast animals increased the percentage of time spent in such high-HR categories as rearing and walking; however, there were no corresponding systematic decreases in the Slow animals. Moreover, the Fast and Slow groups differed significantly with regard to the HRs associated with five behavioral categories. The overall pattern of results suggests that some degree of cardiospecific effect may have been exerted by the operant contingencies.  相似文献

A method has been developed for bringing an aggressive response under stimulus control. It was found that the number of conditioned aggyessive responses was maximal when low-weight target rats were used. The latencies of the conditioned responses toward target animals of 124 gm were significantly lower than those toward animals of 221 and 284 gm. The application of an extinction procedure on the conditioned aggressive response substantially increased the total number of emitted aggressive responses.  相似文献

Backward conditioning of place preference has been obtained in hungry rats when wheel running (the rewarding US) is followed immediately by exposure to a distinctive chamber (the CS). We tested whether such backward conditioning would occur with a 10-min delay. In each of four paired exposures, a period of wheel running (2 or 22 h, in separate experiments) was followed by 10 min in the home cage and then 30 min in the CS chamber. Control rats were put in the CS chamber without wheel running. In other similar experiments, a 0-, 10-, or 30-min delay separated wheel running from exposure to the CS chamber. Reliable conditioned place preference (CPP) occurred when the delay was 0 or 10 but not 30 min. The strength of CPP decreased as the delay increased. These findings imply that the reward process initiated by wheel running remains active for some time after running stops. They support the view that the suppression of feeding produced by wheel running in hungry rats (activity anorexia) is mediated by this reward process.  相似文献

Convergent data suggest dissociated roles for the lateral (LA) and basolateral (BLA) amygdaloid nuclei in fear conditioning, depending on whether a discrete conditioned stimulus (CS)-unconditional stimulus (US) or context-US association is considered. Here, we show that pretraining inactivation of the BLA selectively impaired conditioning to context. In contrast, inactivation of the LA disrupted conditioning to the discrete tone CS, but also either impaired or enhanced contextual conditioning, depending on whether the context was in the foreground or in the background. Hence, these findings refine the current model of the amygdala function in emotional learning by showing that the BLA and the LA not only differentially contribute to elemental and context-US association, but also promote, through their interaction, the most relevant of these two associations.  相似文献

Naive rats were trained to leverpress with a 30-sec delay-of-reinforcement contingency from the start of training. In Experiment 1, the delay interval for different groups of subjects included a signal in the first 5 sec, a signal in the last 5 sec, or no signal at any time. Rats with the signal at the start of the delay interval learned most rapidly. Rats with the signal at the end of the delay failed to learn. In Experiment 2, a choice procedure was used, in which each of two levers was associated with its own 30-sec delay of reinforcement. The delay for one lever included a 5-sec signal at the end of the delay. The delay for the second lever had no signal in its 30-sec delay. Preference was in favor of the lever without the signal in the delay interval. The results demonstrate that the acquisition of new response can be blocked in a manner analogous to the blocking of Pavlovian conditioning.  相似文献
