Scopolamine effects on memory retention in mice: a model of dementia?   总被引:4,自引:0,他引:4  
Scopolamine-treated normal young human subjects exhibit memory dysfunctions analogous to those observed in demented patients. The dysfunctions are reversible by physostigmine but not by d-amphetamine which suggests that the memory impairment is specifically related to reduced cholinergic transmission caused by scopolamine. Scopolamine-induced amnesia has been proposed as a model for dementia where reduced cholinergic function is the suspected cause. We report seven experiments in young adult mice which examine scopolamine's effects on memory retention and whether its amnestic effects are specifically blocked by cholinergic agonists or cholinomimetics. Young adult mice were trained to avoid footshock in a T maze and their retention tested 1 week after training. Pretraining subcutaneous injection of scopolamine improved retention scores of "undertrained" mice at a dose of 0.01 mg/kg but impaired at a dose of 0.1 mg/kg. Post-training injection showed no effect at 0.01 mg/kg, enhanced retention scores at 0.1 mg/kg, and impaired at 1.0 mg/kg. The impairment by 1.0 mg/kg was blocked by injection 45 min post-training of each of two cholinergic drugs but was also counteracted by six drugs which act upon five other neural systems (catecholamine, serotonin, glycine, GABA, and hormonal). When scopolamine was injected 40 min pretraining, and each of eight drugs was injected immediately after training, the amnestic effect of scopolamine was only partially counteracted. This suggests that scopolamine impaired acquisition, in addition to some impairment of memory processing. This was confirmed by a direct study of acquisition rates of the avoidance response; 0.1 mg/kg of scopolamine impaired acquisition. The overall results indicate that pretraining administration of scopolamine impairs learning and to some degree memory processing. Counteracting scopolamine-induced amnesia, by either pretraining or post-training drug administration, is not specific to the cholinergic system.  相似文献

The purpose of this research was to analyze the effects of multimedia training on phonological awareness and word recognition in dyslexic children. We used a control pretest-posttest design, and a sample of 62 children (26 male, 36 female) was selected from Primary Education. Children were selected and classified into two different groups: (1) an experimental group (N=32), and (2) a control group (N=30). The average range age was 9 and 12 years (M=126.7, SD=11.7). We administered phonological awareness tasks, which include different types of syllabic structure from the Sicole-R Multimedia Battery for assessment of cognitive processes in reading. We analyzed whether the Tradislexia videogame affected phonological awareness, considering separately the complexity of syllable structure and type of phonological awareness task. We also analyzed whether the gains in phonological processes were related to training based on type of task or type of syllable structure. The results showed that when we controlled the position of phoneme, the multimedia treatment in segmentation and blending with words that include CV syllables is a better predictor to explain improvement of word decoding processes.  相似文献

The discrepant results noted by the La Trobe/Monash, Sussex, Open, and Berkeley University memory research groups employing the passive avoidance task (PAT) with the chick indicate that some of these differences may be due to differences in training procedures between the various groups. The procedures employed by each group were replicated as closely as possible and compared using the same strain of chick. Higher levels of pecking and lower training latencies were observed in the La Trobe/Monash chicks. Improved training latency was observed following a change in the day of experimentation with Sussex chicks, and in white light in Open chicks. A powerful reinforcing effect was observed when pretraining was conducted with a wet lure across multiple pretraining trials, indicating the importance of this difference between groups. The observed impact on chick performance of these variables indicates that differences between PAT procedures may have important interactive effects with aversive learning. The results of data gathered using different procedures may only be superficially comparable, and further investigation of the effects of these variables is indicated.  相似文献

Sleep may help consolidate the information of certain memories, though its benefits in the consolidation of trace-conditioned memory still remain elusive. We investigated the effect of sleep deprivation on trace learning in male wistar rats. Rats were trained for trace conditioning and the number of head entries into liquid dispenser was accounted as an outcome measure of trace-learning. For training and testing, 75 presentations of conditioned stimulus (CS) (light) and unconditioned stimulus (US) (juice) were offered in five sessions (15 presentations/session; with 5 min inter-session gap). The duration of CS and US stimuli were 15 and 20s respectively, with 5s trace delay between stimuli and 20s condition delay between each presentation. The animals were divided randomly into three groups soon after training, sleep deprived (SD) (n=8), non-SD (NSD) (n=8) and stress control (n=5) groups. The animals of NSD and control groups were left undisturbed, while SD animals were sleep deprived for 6h after training. The learning of trace-conditioned task was examined on following days. We observed that SD rats poked approximately 63% less than NSD and control groups (p<0.001) to obtain juice on testing day. Also, the NSD rats exhibited significant positive correlation in number of head entries during the training and testing days; while the SD rats showed no significant correlation. The results demonstrate that SD animals had difficulties to associate CS with US and suggest that sleep deprivation soon after training impairs the encoding of trace memory.  相似文献

Outservice practicum training of employment service counselors presents a particular challenge to counselor educators. An overview of a two-week practicum-laboratory training program is presented and discussed. The efficacy of the training is examined from the standpoint of helpful levels of empathy presented by the employment service counselors trained. Research on the improvement of elementary school mathematics has shown that computer-based training of number sense (e.g., processing magnitudes or locating numbers on the number line) can lead to substantial achievement gains in arithmetic skills. Recent studies, however, have highlighted that training domain-general cognitive abilities (e.g., working memory [WM]) may also improve mathematical achievement. This study addressed the question of whether a training of domain-specific number sense skills or domain-general WM abilities is more appropriate for improving mathematical abilities in elementary school. Fifty-nine children (Mage = 9 years, 32 girls and 27 boys) received either a computer-based, adaptive training of number sense (n = 20), WM skills (n = 19), or served as a control group (n = 20). The training duration was 20 min per day for 15 days. Before and after training, we measured mathematical ability using a curriculum-based math test, as well as spatial WM. For both training groups, we observed substantial increases in the math posttest compared to the control group (d = .54 for number sense skills training, d = .57 for WM training, respectively). Whereas the number sense group showed significant gains in arithmetical skills, the WM training group exhibited marginally significant gains in word problem solving. However, no training group showed significant posttest gains on the spatial WM task. Results indicate that a short training of either domain-specific or domain-general skills may result in reliable short-term training gains in math performance, although no stable training effects were found in the spatial WM task.  相似文献

Currently, evidence for the beneficial effects of working memory (WM) training on transfer measures in children with attention‐deficit/hyperactivity disorder (ADHD) is inconsistent. Although there is accumulating evidence for the role of individual differences in training and transfer gains of cognitive training, this area has been left unexplored for children with ADHD. In the current study, an advanced latent growth curve model analysis was used to investigate the individual differences in learning curves (training gains) of WM training tasks within a new cognitive intervention ‘Paying Attention in Class’. It was investigated whether certain baseline variables (age, intelligence quotient, externalizing behaviour problems and presence of learning disability) predicted the learning curves and how these individual learning curves influenced near‐transfer and far‐transfer measures. A total of 164 children diagnosed with ADHD, between the age of 8 and 12 years old, followed this new Paying Attention in Class intervention. WM (near‐transfer) and academic performance (far‐transfer) measures were assessed before treatment and directly after treatment. Results showed that individual differences at the start of training were predicted by age and intelligence quotient, but the individual differences in learning curves were not predicted by any of the baseline variables. Both for the verbal and the visuospatial WM training, children with larger training gains (i.e. steeper training curves) showed larger benefits on the near untrained transfer measures. These effects were absent for the far‐transfer measures. Current study shows that training WM is quite complex and has its limitations for children with ADHD. Nonetheless, it highlights that training and transfer gains are affected by many different factors and warrants the need of a more in‐depth investigation of individual differences in future studies.Copyright © 2017 John Wiley & Sons, Ltd.  相似文献

This study evaluated the impact of two interventions—a training program and stimulant medication—on working memory (WM) function in children with attention deficit hyperactivity disorder (ADHD). Twenty‐five children aged between 8 and 11 years participated in training that taxed WM skills to the limit for a minimum of 20 days, and completed other assessments of WM and IQ before and after training, and with and without prescribed drug treatment. While medication significantly improved visuo‐spatial memory performance, training led to substantial gains in all components of WM across untrained tasks. Training gains associated with the central executive persisted over a 6‐month period. IQ scores were unaffected by either intervention. These findings indicate that the WM impairments in children with ADHD can be differentially ameliorated by training and by stimulant medication. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

The role of language in memory for arithmetic facts remains controversial. Here, we examined transfer of memory training for evidence that bilinguals may acquire language-specific memory stores for everyday arithmetic facts. Chinese-English bilingual adults (n = 32) were trained on different subsets of simple addition and multiplication problems. Each operation was trained in one language or the other. The subsequent test phase included all problems with addition and multiplication alternating across trials in two blocks, one in each language. Averaging over training language, the response time (RT) gains for trained problems relative to untrained problems were greater in the trained language than in the untrained language. Subsequent analysis showed that English training produced larger RT gains for trained problems relative to untrained problems in English at test relative to the untrained Chinese language. In contrast, there was no evidence with Chinese training that problem-specific RT gains differed between Chinese and the untrained English language. We propose that training in Chinese promoted a translation strategy for English arithmetic (particularly multiplication) that produced strong cross-language generalization of practice, whereas training in English strengthened relatively weak, English-language arithmetic memories and produced little generalization to Chinese (i.e., English training did not induce an English translation strategy for Chinese language trials). The results support the existence of language-specific strengthening of memory for everyday arithmetic facts.  相似文献

Expert team-handball players do not show the typical trade-off between speed and accuracy in overarm throwing. Van den Tillaar and Ettema (2003a) attributed this result to the uniqueness of the training experience of this group. The purpose of this study was to test their hypothesis by comparing experts with novices on overarm throwing by manipulating the goal of the task using different instructions. No trade-off between speed and accuracy was found for novices (n = 13; M age = 22.7 yr., SD = 2.2) or experts (n = 9; Mage = 24 yr., SD = 2.2): accuracy did not change by instruction. Furthermore, the linear velocities of the ball and endpoints of body segments and their timing were affected by instruction in a similar fashion in both groups. This finding indicates that training experience is not related to speed-accuracy tradeoff in overarm throwing.  相似文献

Adolescent rats are less sensitive than adults to the hypothermic, anxiolytic, motor impairing, hypnotic, and lethal effects of ethanol. In vitro experiments nevertheless suggest that hippocampal neural activity is more affected by ethanol in preweanling or adolescent rats than in adults. These data are complemented by in vivo results showing that pretraining ethanol impairs learning in adolescent rats at doses that do not affect adult learning. In order to determine if posttraining ethanol affects memory differently in adolescents than in adults, Sprague-Dawley albino rats of both ages were trained in an appetitively motivated odor discrimination in which they were required to dig in scented sand for sweetened cereal reward. Immediately after training subjects received intraperitoneal injections of 0, 0.5, or 1g/kg ethanol (12.6%). At test, 48h later, subjects were presented with unbaited discriminanda and the time (s) spent digging in the S+ and S- was measured. Adolescents, but not adults, showed impaired discrimination performance if training was followed by ethanol. A subsequent experiment discounted the possibility that impaired adolescent performance was due to ethanol-induced conditioned taste or odor aversions. It thus appears that relative to adults, memory in adolescent rats is more strongly affected by ethanol in a test of appetitive conditioning that excludes ethanol's effects on sensory and motivational influences during the learning experience.  相似文献

Traditional approaches in stimulating moral growth following Turiel's (1966) "+1" intervention strategy have been only marginally successful. This failure rate may be due to (a) the inconsistent control of both the cognitive and social perspective-taking prerequisites to moral growth, and (b) an overestimation of subjects' interpersonal communication skills. In this study, 17 college undergraduates (8 males, 9 females), possessing either basic or consolidated formal operations, received eight weeks of interpersonal skills training, along with pre- and posttraining assessments of social perspective-taking and moral reasoning. It was expected that gains in moral reasoning would be related to subjects' possessing the prerequisite cognitive and perspective-taking competencies necessary for moral growth. Results showed that posttraining changes in moral reasoning were in the predicted direction. In addition, gender differences in social perspective-taking were evident; males had significantly lower social perspective-taking scores at pretraining and showed significant gains in perspective-taking at posttesting. Females, however, scored relatively high in social perspective-taking at pretraining and showed no significant change in ability at posttesting. The results are interpreted as supportive of existing theoretical models regarding these domains of development.  相似文献

In the current study, we examined the degree to which sudden gains (large, rapid, and stable symptom reduction in a one-session interval) predicted treatment outcome in adults randomized to two different trauma-focused treatments. Adults diagnosed with PTSD were randomized to either written exposure therapy (WET; n = 63), a brief, exposure-based treatment for posttraumatic stress disorder (PTSD), or the more time-intensive Cognitive Processing Therapy (CPT; n = 63). Findings showed that 20.6% of participants who received WET and 17.5% of participants who received CPT experienced sudden gains. Sudden gains occurred earlier in WET (M session = 2.69, SD = 0.75) than in CPT (M session = 5.64, SD = 3.01). However, there were no treatment condition differences in the magnitude of the sudden gains. Treatment outcomes were significantly better for those who experienced sudden gains compared with those who did not, regardless of treatment assignment. Exploratory analyses of participants’ trauma narratives revealed that expressing more negative emotion predicted the occurrence of sudden gains in both treatment conditions. Negative beliefs about the self and others did not predict sudden gains. The findings are discussed in terms of how they may help identify individual early response patterns that predict outcomes in trauma-focused treatments.  相似文献

The effect on the capacity of visual working memory of spatial complexity (as defined by Garner's principle) in rotation and reflection transformation was examined in persons differing along the Autism Spectrum Quotient (AQ), using a change-detection task. On each trial, nine line segments were arrayed in simple, medium, and complex configurations, which were presented in memory and test displays. 27 participants (8 men, 19 women; M age = 22.3 yr., SD = 2.7) were asked whether the orientations of stimuli between two displays were the same or different. On the basis of their AQ scores out of 50 (M AQ scores = 20.9, SD = 6.3), the participants were divided into groups with high (n = 13; M AQ scores = 26.2, SD = 4.1) and low (n = 12; M AQ scores = 15.3, SD = 2.7) self-reported autistic-like traits (High and Low AQ groups, 2 excluded for scores at the median). The results showed that spatial complexity affects the capacity of visual working memory for the Low AQ group but not for the High AQ group, suggesting the functional dissociation of spatial configuration and visual working memory in the High AQ group.  相似文献

A preseason mental skills program for serving was implemented for the 11 members of an intercollegiate volleyball team (M age = 20.0 yr.; SD = 1.1; years of intercollegiate volleyball experience M = 2.6; SD = 0.9). Key mental skills taught were relaxation, imagery, attentional focus, goal setting, behavioral modeling, and performance routine. A videotaped behavioral model articulated and demonstrated technical performance keys to effective serving. Players utilized a three-phase service routine to increase automaticity of performance and to incorporate key mental skills. End-of-season reported use of imagery was significantly correlated with Good Serve Percentage, as was reported use of a service routine. The mean Good Serve Percentage for the season was 49% (SD=7); the team goal was 50%. Serve-specific self-efficacy significantly increased from the pretraining program to the end of the season. Results indicated that implementing the mental skills training program was associated with enhanced service performance.  相似文献

The cytosolic posttranslational protein-modifying mechanism of monoADP-ribosylation has been implicated in long-term potentiation, a synaptic model of memory formation. The current study investigated the effect of inhibiting mono(ADP-ribosyl) transferase on memory for the passive avoidance task in day-old chicks (white Leghorn-black Australorp). Various doses of novobiocin or menadione sodium bisulfite were administered intracranially at different times before or after training. Control chicks were administered saline at matched times. Novobiocin (650 microM) or menadione sodium bisulfite (250 microM) administered between 5.0 min pretraining and 2.5 min posttraining was found to cause a persistent loss of retention from 120 min posttraining. These data provide the first demonstration that monoADP-ribosylation is required for the maintenance of long-term memory. Furthermore, the temporal characteristics of the memory loss caused by monoADP-ribosylation inhibition appears to exclude this mechanism as a downstream effect of the well-established nitric oxide activity previously shown to occur within 40 min of passive avoidance training.  相似文献

The current study reports on an intervention study that investigated the relative contributions of 5‐week English language training (ELT) and 5‐week working memory training (WMT) to improvements in English language knowledge and working memory capacity in university learners of English as a foreign language. The participants were allocated to one of the following three experimental groups and a control group: (i) ELT only; (ii) WMT only; and (iii) both WMT and ELT. The results showed that intervention effects were pronounced in the WMT + ELT group, who improved scores on the reading span test and all tests of verbal and visuo‐spatial short‐term and working memory. The gains on verbal short‐term (Japanese) and working memory (Japanese and English) were maintained 3 months later. None of the interventions, single or combined, were associated with gains in foreign language knowledge, however. The sustained effects of the dual approach and its implications are discussed. Extensive evidence indicates that epinephrine (EPI) modulates memory consolidation for emotionally arousing tasks in animals and human subjects. However, previous studies have not examined the effects of EPI on consolidation of recognition memory. Here we report that systemic administration of EPI enhances consolidation of memory for a novel object recognition (NOR) task under different training conditions. Control male rats given a systemic injection of saline (0.9% NaCl) immediately after NOR training showed significant memory retention when tested at 1.5 or 24, but not 96h after training. In contrast, rats given a post-training injection of EPI showed significant retention of NOR at all delays. In a second experiment using a different training condition, rats treated with EPI, but not SAL-treated animals, showed significant NOR retention at both 1.5 and 24-h delays. We next showed that the EPI-induced enhancement of retention tested at 96h after training was prevented by pretraining systemic administration of the beta-adrenoceptor antagonist propranolol. The findings suggest that, as previously observed in experiments using aversively motivated tasks, epinephrine modulates consolidation of recognition memory and that the effects require activation of beta-adrenoceptors.  相似文献

Research has established that challenging memory goals always lead to score increases for younger adults, and can increase older adults' scores under supportive conditions. This study examined beliefs and on-task effort as potential mechanisms for these self-regulatory gains, in particular to learn whether episodic memory gains across multiple trials of shopping list recall are controlled by the same factors for young and old people. Goals with feedback led to higher recall and strategic categorisation than a control condition. Strategy usage was the strongest predictor of gains over trials for both age groups. Age, goal condition, and effort also predicted scores across the entire sample. Older adults' gains, but not younger adults' gains, were affected significantly by the interaction of self-efficacy beliefs and goal condition, and condition interacted with locus of control to predict younger adult gains. These results emphasise the importance of self-regulatory effort and positive beliefs for facilitating goal-related memory gains.  相似文献

Fluid intelligence (Gf) predicts performance on a wide range of cognitive activities, and children with impaired Gf often experience academic difficulties. Previous attempts to improve Gf have been hampered by poor control conditions and single outcome measures. It is thus still an open question whether Gf can be improved by training. This study included 4-year-old children (N = 101) who performed computerized training (15 min/day for 25 days) of either non-verbal reasoning, working memory, a combination of both, or a placebo version of the combined training. Compared to the placebo group, the non-verbal reasoning training group improved significantly on Gf when analysed as a latent variable of several reasoning tasks. Smaller gains on problem solving tests were seen in the combination training group. The group training working memory improved on measures of working memory, but not on problem solving tests. This study shows that it is possible to improve Gf with training, which could have implications for early interventions in children.  相似文献
